Advances in Statistical Network Modeling and Nonlinear Time Series Modeling [PDF]
The thesis is composed of two independent topics: statistical network modeling and nonlinear time series modeling. With the increasing demand of network data analysis, we present two statistical network models and inferences, analyzing both theoretical ...

Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.

Keratin 19 (KRT19) is overexpressed in high‐grade serous ovarian cancer with high levels of Kallikrein‐related peptidases (KLK) 4–7 and is associated with poor survival. In vivo analyses demonstrate that elevated KRT19 increases peritoneal tumour burden.

Targeted therapy was evaluated in SHH medulloblastoma using neuroepithelial stem cell (NES) and tumor‐derived NES‐like (tNES) models in 2D monolayers and 3D spheroids. PI3K, AKT, and CDK4/6 inhibitors had minimal effects in NES but markedly reduced viability and growth and induced apoptosis in tNES cells, revealing distinct therapeutic vulnerabilities.

We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...

Spectral expansion methods for prediction uncertainty quantification in systems biology
Uncertainty is ubiquitous in biological systems. For example, since gene expression is intrinsically governed by noise, nature shows a fascinating degree of variability.

Pancreatic sensory neurons innervating healthy and PDAC tissue were retrogradely labeled and profiled by single‐cell RNA sequencing. Tumor‐associated innervation showed a dominant neurofilament‐positive subtype, altered mitochondrial gene signatures, and reduced non‐peptidergic neurons.

Modeling the dynamics of anti-microbial resistant cholera infection with reinfection
Cholera continues to pose a significant global public health challenge. The occurrence of severe weather events, including floods, droughts, and cyclones, has led to a surge in new cholera outbreaks and exacerbated both epidemic and endemic conditions in
