Perampanel Blocks Transsynaptic α‐Synuclein Propagation and Neurodegeneration in a Mouse Model of Lewy Body Disease

Abstract Background Transsynaptic α‐synuclein propagation plays a crucial role in the progression of Lewy body disease. We previously demonstrated that an α‐amino‐3‐hydroxy‐5‐methyl‐4‐isoxazolepropionic acid receptor antagonist, perampanel, blocks neuronal uptake of α‐synuclein preformed fibrils (PFFs) in an activity‐dependent manner.

Tau‐targeting active immunotherapy slows progression and reduces pathology in mouse models of tauopathy

The efficacy of the novel anti‐tau active immunotherapy, p5555kb, was tested using two mouse models of tau pathology. p5555kb inoculation increased the survival rate and reduced tau pathology in tau‐overexpressing P301L mice and decreased tau seeding in the brains of C57BL/6 mice injected with human‐purified Alzheimer's disease tau.

Hcfc1 and Ogt Mediate Zebrafish CNS Regeneration Through Hippo/Yap Signalling

This work identifies Hcfc1 and Ogt as key regulators of zebrafish CNS regeneration through modulation of Hippo/Yap signalling. Loss of Hcfc1 or Ogt activity inhibition impairs regeneration, which is restored by Yap overexpression, revealing a new regulatory axis that enhances CNS regenerative capacity.

Lactate Accelerates Early Angiogenesis and Bone Regeneration Through Macrophage M1 Polarisation

During the early stage of bone defect healing, lactate accumulates and contributes to increasing NOD1 expression by stabilising HIF1α that in turn triggers a calcium influx, which ultimately polarises macrophages towards the M1 phenotype and accelerates vascularisation of endothelial cells.

Abstract Objective Accumulating evidence indicates that aberrant C1q‐C3 complement signaling in microglia and astrocytes drives synaptic dysfunction and neuronal loss. C1q‐mediated synaptic dysfunction disrupts neuronal circuitry balance and can lead to network hyperexcitability in epilepsy.

Daily intermittent fasting is an effective multiscale treatment in preclinical models of absence epilepsy
