Long‐read sequencing‐based atlas of tissue‐specific expression of DNM1L transcript variants

open access: yesThe FEBS Journal, EarlyView.
Targeted long‐read sequencing resolves full‐length DNM1L (Drp1) isoforms and reveals conserved, tissue‐specific expression patterns across human and mouse tissues. Functional assays show that Drp1 isoforms differ in their ability to drive mitochondrial fission, independent of abundance, with specific exons modulating activity.

Structural insights into θ‐type carbonic anhydrases 3 and 4: Tuning the directionality of CO2 hydration in a diatom

open access: yesThe FEBS Journal, EarlyView.
θ‐CA3 from a marine diatom has an N‐terminal transmembrane helix and two homologous catalytic domains. The enzyme functions as a dimer, with active sites formed near the dimer interface. A bicarbonate ion binds on the symmetry axis of the dimer and forms hydrogen bonds with both monomers, thereby activating θ‐CA3 and tuning CO2 hydration activity ...

The trimeric Shu complex in C. elegans is an ATPase that remodels RAD51 filaments in the homologous recombination‐associated DNA damage response

open access: yesThe FEBS Journal, EarlyView.
The C. elegans Shu complex (RFS‐1/RIP‐1/SWS‐1) is a DNA‐dependent ATPase that regulates RAD‐51 filaments during homologous recombination. The trimer preferentially binds 5′‐exposed DNA, remodels RAD‐51 filaments via ATP hydrolysis, and stabilizes filaments through ATP binding.

Mechanisms underpinning the progression of nondysplastic Barrett's oesophagus to cancer

open access: yesThe FEBS Journal, EarlyView.
Barrett's oesophagus confers a low but significant cancer risk. The condition is common but we do not have effective biomarkers to identify susceptible patients. We believe that there is no single factor that determines cancer risk and an understanding of the combinations of interactions (via gene expression and natural selection) between the Barrett's

Newly identified human aminoacyl‐tRNA synthetase complex interacting multifunctional protein 2 (AIMP2) loss‐of‐function mutations cause neurodevelopmental defects linked to cell death in a zebrafish model

open access: yesThe FEBS Journal, EarlyView.
Human AIMP2 mutations lead to severe neurodevelopmental defects and brain atrophy. Using patient‐derived fibroblasts from two individuals, we show decreased AIMP2 protein levels and overall protein synthesis. In a zebrafish loss‐of‐function model, the lack of AIMP2 leads to an increase in cell death and results in smaller brains.

Combining TAG‐72CAR with anti‐MUC16CAR improves the potency of ovarian cancer treatment

open access: yesThe FEBS Journal, EarlyView.
Tz‐KBBCAR, a combinatorial CAR targeting MUC16 and TAG‐72, enhances the precision of ovarian cancer immunotherapy. By requiring both tumour antigens for full activation, Tz‐KBBCAR was designed to eliminate MUC16+/TAG‐72+ cancer cells, while sparing MUC16+/TAG‐72‐ healthy cells.
