Expanding Genetic Code to Generate Human Brain Organoids with Both Vasculature and Microglia‐Like Cells

Using genetic code expansion, we engineered vascularized human cerebral organoids (vhCOs) with microglia‐like cells and blood‐brain barrier features. vhCOs recapitulate neurovascular interactions, regional identities, and neuronal subtypes resembling the fetal brain.

Collagen‐Binding IL‐15 Immunocytokine Armed Oncolytic Adenovirus Establishes a Self‐Reinforcing Immunostimulatory Milieu to Potentiate Antitumor Immunity

Repurposing the dense tumor stroma from a physical barrier into an anchoring scaffold, an engineered oncolytic adenovirus encoding IL15C‐CBD overcomes the immunosuppressive microenvironment in triple‐negative breast cancer. Intratumoral viral replication secretes this collagen‐anchored immunocytokine, forming a localized cytokine depot.

Concurrent YAP hyperactivation and Tp53 loss in Atp4b‐expressed parietal cell lineage drives gastric cancer initiation, multi‐organ metastasis, and intrinsic resistance to anti‐PD‐1 therapy. BST2 emerges as a direct YAP‐TEAD transcriptional target that engages PIRA2 on myeloid cells to establish an immunosuppressive microenvironment.
