Artifactual detection of biotin on histones by streptavidin
Analytical Biochemistry, 2008Biotinylation is a recent addition to the list of reported posttranslational modifications made to histones. Holocarboxylase synthetase (HCS) and biotinidase have been implicated as biotinylating enzymes. However, the details of the mechanism and the regulation of biotin transfer on and off histones remains unclear.

Molecular Determinants and Bottlenecks in the Dissociation Dynamics of Biotin-Streptavidin.
Journal of Physical Chemistry B, 2017Biotin-streptavidin is a very popular system used to gain insight into protein-ligand interactions. In its tetrameric form, it is well-known for its exceptionally high kinetic stability, being one of the strongest known noncovalent interactions in nature,

The goal of this project was to express streptavidin on the outer surface of Escherichia coli so that we could target any biotinylated nucleic acid or peptide to a cell surface. We used BioBrick assembly to fuse together a lipoprotein signal peptide, outer membrane protein A transmembrane domains and streptavidin protein domains. Assembly of constructs

Quantification of streptavidin adsorption in microtitration wells
Analytical Biochemistry, 2004Streptavidin-coated microtitration plates have an important role as a solid phase in clinical diagnostics. We have designed techniques for evaluating quantitative and functional aspects of streptavidin adsorbed in microtitration wells. The theoretical monolayer adsorption capacity was modeled based on the molecular dimensions of the protein.

Artificial ligands of streptavidin (ALiS) with association constants of ∼10(6) M(-1) were discovered by high-throughput screening of our chemical library, and their binding characteristics, including X-ray crystal structure of the streptavidin complex ...

An artificial imine reductase results upon incorporation of a biotinylated Cp*Ir moiety (Cp* = C5Me5(-)) within homotetrameric streptavidin (Sav) (referred to as Cp*Ir(Biot-p-L)Cl] ⊂ Sav).

Streptavidin-reactive protein in Fasciola hepatica
Veterinary Parasitology, 1995Streptavidin-binding protein was found in an extract of Fasciola hepatica, and the protein was analyzed by the inhibition assay using avidin or D-biotin, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blotting using peroxidase-conjugated streptavidin.

Conjugation of the Photoluminescent Silicon Nanoparticles to Streptavidin
Bioconjugate Chemistry, 2008We have covalently attached multiple photoluminescent silicon nanoparticles (SNs) to streptavidin molecules. Conjugation of SNs to a target protein is achieved using the multistage photoassisted procedure. In a first step, the terminal hydrogen in the freshly prepared SNs is substituted with an alkane monolayer that serves as a platform for chemical ...

Structural origins of high-affinity biotin binding to streptavidin.
Science, 1989The high affinity of the noncovalent interaction between biotin and streptavidin forms the basis for many diagnostic assays that require the formation of an irreversible and specific linkage between biological macromolecules.
