Progress in the Development of Subunit Vaccines against Malaria [PDF]

open access: yesVaccines, 2020
Malaria is a life-threatening disease and one of the main causes of morbidity and mortality in the human population. The disease also results in a major socio-economic burden. The rapid spread of malaria epidemics in developing countries is exacerbated by the rise in drug-resistant parasites and insecticide-resistant mosquitoes.

Enhanced Intracellular Stability and Translation Efficiency of mRNA Drugs by a 2‐arm mRNA Platform

open access: yesAdvanced Science, EarlyView.
We constructed a 2‐arm mRNA, characterized by a unique topology formed through the dimerization of two mRNA 3’ tails. The 2‐arm mRNA improves 3’ tail stability and resistance to nuclease degradation, resulting in an intracellular half‐life of up to 65 h. This method substantially enhances the translation capacity of mRNA drugs.

BCMA‐Engineered Dendritic Cell‐Derived Exosomes as Bi‐Functional Therapeutics Orchestrating Cytokine Sequestration and Immune Activation for Multiple Myeloma

open access: yesAdvanced Science, EarlyView.
A dual‐function cell‐free therapeutic based on DC2.4 cell‐derived exosomes engineered to display BCMA. (Left) Soluble Ligand Sequestration (Decoy Function): DB Exo act as molecular decoys that predominantly sequester soluble APRIL with partial BAFF attenuation, effectively disrupting the NF‐κB survival signaling axis and suppressing myeloma cell ...

Maternal and protective antibody responses to E2 subunit and C-strain classical swine fever vaccines in piglets: a comparative analysis

open access: yesFrontiers in Veterinary Science
Classical swine fever (CSF), caused by the classical swine fever virus (CSFV), is an acute and highly contagious disease affecting swine. In clinical settings, the C- strain vaccine and the E2 subunit vaccine are commonly employed for the prevention and ...

Advancements in the development of subunit influenza vaccines [PDF]

open access: yesMicrobes and Infection, 2015
The ongoing threat of influenza epidemics and pandemics has emphasized the importance of developing safe and effective vaccines against infections from divergent influenza viruses. In this review, we first introduce the structure and life cycle of influenza A viruses, describing major influenza A virus-caused pandemics.

Senescent Cell Derived Artificial Vesicle‐Based Senolytic Sonovaccine Platform with Augmented Lymph Node Delivery and Antigen Cross‐Presentation Efficacy

open access: yesAdvanced Science, EarlyView.
A senolytic sonovaccine platform (SenoVac) is developed, in which senescent cell‐derived vesicles serve as broad senescent cell antigen reservior. The “2‐step” click chemistry strategy for effective lymph node delivery, and ultrasound‐triggered endosomal escape to boost cross‐presentation, ensures efficient senescent cell clearance and disease ...

Introduction of neutralizing immunogenicity index to the rational design of MERS coronavirus subunit vaccines

open access: yesNature Communications, 2016
Viral subunit vaccines contain epitopes that elicit non-neutralizing immune responses. Here, Duet al. mask an immunodominant non-neutralizing epitope on a MERS coronavirus subunit vaccine with a glycan probe, leading to significantly improved efficacy of

ProSiteHunter: A Unified Framework for Sequence‐Based Prediction of Protein‐Nucleic Acid and Protein‐Protein Binding Sites

open access: yesAdvanced Science, EarlyView.
This study proposed a unified sequence‐based framework for protein binding site prediction, which adopted a tri‐track semantic multi‐source feature fusion strategy to effectively capture diverse macromolecular interaction sites and further improved the accuracy of antibody‐antigen interaction prediction.

Impact of adjuvant: Trivalent vaccine with quadrivalent-like protection against heterologous Yamagata-lineage influenza B virus

open access: yesFrontiers in Immunology, 2022
As new vaccine technologies and platforms, such as nanoparticles and novel adjuvants, are developed to aid in the establishment of a universal influenza vaccine, studying traditional influenza split/subunit vaccines should not be overlooked. Commercially

Delivery of STING agonists for adjuvanting subunit vaccines

open access: yesAdvanced Drug Delivery Reviews, 2021
Adjuvant is an essential component in subunit vaccines. Many agonists of pathogen recognition receptors have been developed as potent adjuvants to optimize the immunogenicity and efficacy of vaccines. Recently discovered cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway has attracted much attention as it is a key mediator for ...
