In situ molecular organization and heterogeneity of the Legionella Dot/Icm T4SS

open access: yesFEBS Letters, EarlyView.
We present a nearly complete in situ model of the Legionella Dot/Icm type IV secretion system, revealing its central secretion channel and identifying new components. Using cryo‐electron tomography with AI‐based modeling, our work highlights the structure, variability, and mechanism of this complex nanomachine, advancing understanding of bacterial ...

Polynomial Chaos-based Bayesian Inference of K-Profile Parametrization in a General Circulation Model of the Tropical Pacific

open access: yes, 2015
The authors present a Polynomial Chaos (PC)-based Bayesian inference method for quantifying the uncertainties of the K-Profile Parametrization (KPP) within the MIT General Circulation Model (MITgcm) of the tropical pacific. The inference of the uncertain

Cell wall target fragment discovery using a low‐cost, minimal fragment library

open access: yesFEBS Letters, EarlyView.
LoCoFrag100 is a fragment library made up of 100 different compounds. Similarity between the fragments is minimized and 10 different fragments are mixed into a single cocktail, which is soaked to protein crystals. These crystals are analysed by X‐ray crystallography, revealing the binding modes of the bound fragment ligands.

AI-Driven Surrogate Model for Room Ventilation

open access: yesFluids
The control of ventilation systems is often performed by automatic algorithms which often do not consider the future evolution of the system in its control politics. Digital twins allow system forecasting for a more sophisticated control.

Decomposition-Assisted Computational Technique Based on Surrogate Modeling for Real-Time Simulations

open access: yesComplexity, 2017
The development of complex simulation systems is extremely costly as it requires high computational capability and expensive hardware. As cost is one of the main issues in developing simulation components, achieving real-time simulation is challenging ...

The (Glg)ABCs of cyanobacteria: modelling of glycogen synthesis and functional divergence of glycogen synthases in Synechocystis sp. PCC 6803

open access: yesFEBS Letters, EarlyView.
We reconstituted Synechocystis glycogen synthesis in vitro from purified enzymes and showed that two GlgA isoenzymes produce glycogen with different architectures: GlgA1 yields denser, highly branched glycogen, whereas GlgA2 synthesizes longer, less‐branched chains.

Online learning supported surrogate-based flowsheet model maintenance

open access: yesDigital Chemical Engineering
Surrogate-based flowsheet model calibration is a critical extension of using flowsheet models in Digital Twin (DT) systems. However, maintaining accurate surrogates over time is increasingly challenging, especially when models are deployed in real-time ...

Structural biology of ferritin nanocages

open access: yesFEBS Letters, EarlyView.
Ferritin is a conserved iron‐storage protein that sequesters iron as a ferric mineral core within a nanocage, protecting cells from oxidative damage and maintaining iron homeostasis. This review discusses ferritin biology, structure, and function, and highlights recent cryo‐EM studies revealing mechanisms of ferritinophagy, cellular iron uptake, and ...

Surrogate Modeling: Review and Opportunities for Expert Knowledge Integration

open access: yesProcedia Computer Science
Digital Twins (DTs), essential for digitalization and Industry 4.0, demand high-fidelity yet computationally efficient models for real-time replication of physical systems. However, simulating continuous dynamical systems, often stiff and highly granular, poses significant computational challenges.
