Lactate‐Driven Restriction of Mitochondrial Permeability Transition Promotes Resistance to Chemo‐Immunotherapy by Suppressing Tumor PANoptosis

open access: yesAdvanced Science, EarlyView.
Tumor‐derived lactate establishes a metabolic lock that drives chemo‐immunotherapy resistance by suppressing PANoptosis. Mechanistically, KAT8 lactylates the mitochondrial translocator ANT2, which recruits PGAM5 to dephosphorylate CypD. This cascade restricts mitochondrial permeability transition pore opening, preventing pro‐inflammatory mtDNA leakage.

Targeting the ATX‐LPA Axis Overcomes TKI Resistance and Immunosuppression in Renal Cell Carcinoma via Dual Inhibition of AKT/mTOR and TBK1/IRF3 Pathways

open access: yesAdvanced Science, EarlyView.
ABSTRACT Background Therapeutic resistance limits durable survival in advanced/metastatic renal cell carcinoma (RCC) treated with first‐line tyrosine kinase inhibitor (TKI) plus immune checkpoint inhibitor (ICI). We sought to define key resistance drivers and actionable targets.

Multiplex metagenomic sequencing for rapid viral pathogen identification and surveillance in clinical specimens

open access: yesBMC Infectious Diseases
Abstract Background Rapid and accurate viral detection is essential for clinical diagnosis and effective outbreak surveillance. Traditional methods, including culture-based isolation and antigen tests, are time-consuming and limited by tissue tropism.

Calcineurin‐Dependent Stress Adaptation Enables Caspofungin Heteroresistance Leading to Stable Resistance in Candida Glabrata

open access: yesAdvanced Science, EarlyView.
Caspofungin heteroresistance is prevalent in clinical Candida glabrata isolates and depends on calcineurin‐mediated stress adaptation. This transient phenotype serves as a reservoir for resistance evolution, enabling the emergence of stable resistant descendants under prolonged drug pressure.

Targeting p21‐High Senescent Kupffer Cells Nanotherapeutically Potentiates Antitumor Immunity in Advanced Hepatocellular Carcinoma with Portal Vein Tumor Thrombus

open access: yesAdvanced Science, EarlyView.
Schematic illustration of a biomimetic SKEV@AAV platform targeting p21‐high senescent Kupffer cells (sKCs) in hepatocellular carcinoma (HCC) with portal vein tumor thrombus (PVTT). p21‐high sKCs accumulate in the PVTT microenvironment and establish an immunosuppressive, SASP‐rich environment that facilitates tumor cell proliferation and immune evasion.

Radiation embrittlement of reactor pressure vessel materials of Rivne NPP unit 1 due to re-irradiation after recovery annealing

open access: yesЯдерна фізика та енергетика, 2019
The results of surveillance-specimens testing of re-irradiated after recovery annealing reactor pressure vessel (RPV) metal of Rivne NPP unit 1 are presented. The radiation strengthening and shift of critical temperature of brittleness for RPV base metal

Genomic Variant Surveillance of SARS-CoV-2 Positive Specimens Using a Direct PCR Product Sequencing Surveillance (DPPSS) Method

open access: yesActa Medica Philippina
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) as the causative agent of COVID-19 has significantly challenged the public health landscape in late 2019. After almost 3 years of the first ever SARS-CoV-2 case, the World Health Organization (WHO) declared the end of this global health emergency in May 2023.

Multi‐Omics Profiling of High‐Grade Serous Ovarian Cancer Reveals an Inflammation‐Related Lipid Metabolism Subtype Associated With Platinum Resistance

open access: yesAdvanced Science, EarlyView.
Through integrated proteomic and metabolomic profiling, Zhao et al. identified three molecular subtypes of high‐grade serous ovarian cancer. The high‐risk subtype exhibits activated arachidonic acid metabolism and cyclooxygenase‐2 overexpression. This metabolic axis promotes M2‐like macrophage infiltration, which contributes to platinum resistance ...

Rewiring Pyroptosis to Potentiate Cancer Immunotherapy via a Gasdermin D Agonist Bypassing Caspase‐3

open access: yesAdvanced Science, EarlyView.
This work introduces a small‐molecule agonist, DIBDO, that bypasses Casp‐3 to directly trigger GSDMD‐mediated pyroptosis in tumors. Activated by glutathione, DIBDO generates singlet oxygen and molecular iodine, blocking Casp‐3 pathways and inducing immunogenic cell death.
