Minor Taxoids from Taxus wallichiana

Journal of Natural Products, 1995
The needles of Taxus wallichiana afforded a new analogue of taxinine M [1a] and two derivatives of brevifoliol [2a, 3a]. The conformation of 3a was investigated by nmr spectroscopy with the aid of variable-temperature experiments and in situ reactions.

The Chemistry of Taxol and Related Taxoids

2002
The anticancer compound taxol (1.1.1) was originally isolated in the late 1960’s from the bark of the Western Yew, Taxus brevifolia, and its structure was published in 1971 in a paper that has been cited well over 1000 times since its publication (1). Taxol was originally isolated on the basis of its cytotoxicity and antileukemic activity, but it was ...

Synthesis of new taxoids

Tetrahedron, 1994
Abstract [2+2] Photocycloadditions between enone 2 and 2,3-dihydropyran derivatives lead to a number of interesting intermediates 3a–e for taxoid ...

The scientific rationale for developing taxoids

Anti-Cancer Drugs, 1996
The taxoid drugs, docetaxel (Taxotere) and paclitaxel (Taxol), represent a new class of antitumour agents which act by promoting the assembly and inhibiting the disassembly of microtubules. Docetaxel has been shown to be more potent than paclitaxel with regard to the formation and stabilization of microtubules in vitro. Docetaxel also has a higher cell

Preclinical Evaluation of New Taxoids

Current Pharmaceutical Design, 2001
Paclitaxel and docetaxel are two key molecules in the treatment of a variety of cancers with major impact in the treatment of breast, lung and ovarian cancers. A number of taxoids have then been synthesized in an effort to improve some of the features of the existing drugs.

Syntheses and cytotoxicities of the analogues of the taxoid brevifoliol

European Journal of Medicinal Chemistry, 2008
Seven novel brevifoliol analogues have been synthesized by coupling brevifoliol and 2-monosubstituted-4-phenyl-1,3-oxazolidine carboxylic acid after removal of the protecting group with acid treatment. Brevifoliol and its synthesized analogues were tested for their cytotoxic activities against four different human cancer cell lines, oral (KB), breast ...

The synthesis of novel taxoids for oral administration

Bioorganic & Medicinal Chemistry, 2014
A group of novel taxoids, with modifications at C-7, C-10, C-3' and C-14 positions of paclitaxel, was synthesized in order to improve their biological profile by decreasing their affinity with P-glycoprotein (P-gp) and increasing cellular permeability.

Fluorescent taxoids as probes of the microtubule cytoskeleton

Cell Motility and the Cytoskeleton, 1998
Microtubules are specifically and efficiently visualized with the new fluorescent taxoids 7-O-[N-(4'-fluoresceincarbonyl)-L-alanyl]taxol (FLUTAX) and 7-O-[N-(4'-tetramethylrhodaminecarbonyl)-L-alanyl]taxol (ROTAX). Similarly to taxol, FLUTAX and ROTAX are able to drive inactive GDP-liganded tubulin into microtubule assembly.

Studies towards the total synthesis of taxoids. A straightforward procedure for the taxoid BC substructure

Tetrahedron Letters, 1993
Abstract The aldol condensation of the ketone 6 and the aldehyde 7 followed by an annelation-fragmentation afforded the bicyclo[6.4.0]dodecane derivative 3 with the appropriate functionalities for further elaboration.
