SMAD4 Palmitoylation Drives a Metabolic‐Transcriptional Circuit to Promote Tumorigenesis and Confers Radiosensitivity in Pancreatic Cancer

This study identifies palmitoylation as a novel regulatory modification of SMAD4, mediated by ZDHHC22/APT2. It activates fatty acid synthesis, creating a self‐reinforcing SMAD4–FASN–palmitate feedback loop that drives pancreatic cancer growth and enhances radiotherapy sensitivity.

In the present study, we have demonstrated that the NUP85‐USP47‐ASK1 signaling pathway may have regulated the progression of liver fibrosis through modulating ERS. MoHrd3 plays a crucial role in regulating the pathogenicity of Magnaporthe oryzae. As an ER‐associated degradation component, MoHrd3 involves in ER stress‐triggered autophagy. It facilitates the fusion between the autophagosome and the vacuole via enhancing the interaction between MoAtg8 and MoYpt7.

We developed the enzyme‐responsive peptide SAp‐CD28 to selectively target CD28‐overexpressing T‐ALL cells. Following phosphatase‐mediated activation, SAp‐CD28 undergoes conformational switching and nanooligomerization, resulting in the disruption of CD28 downstream signaling.
