Archaebacterial bipolar tetraether lipids: Physico-chemical and membrane properties

Bipolar tetraether lipids (BTL) are abundant in archaea and can be chemically synthesized. The structures of BTL are distinctly different from the lipids found in bacteria and eukaryotes. In aqueous solution, BTL can form extraordinarily stable liposomes with different sizes, lamellarities and membrane packing densities.

A soil profile from the Saxnäs Mosse peat bog, Sweden, has been analysed for glycerol dialkyl glycerol tetraether (GDGT) membrane lipids and 16S rRNA genes in order to constrain the source of the yet ‘orphan,’ but supposedly bacterial, branched GDGTs. Branched GDGT lipids dominate over archaeal membrane lipids.

Unusual Thermal Stability of Liposomes Made from Bipolar Tetraether Lipids

The thermal stability of liposomes made from polar lipids of Sulfolobus acidocaldarius, a thermoacidophilic archaebacterium, was studied. While two analogous nonarchaebacterial liposomes examined were unstable above 50 degrees C, liposomes made of the native tetraether lipids showed stability up to at least 80 degrees C.
