Potentiation effects of the positive allosteric modulator PTC‐174 on NMDA receptors in neonatal male rat substantia nigra dopaminergic neurons

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Background and Purpose Positive allosteric modulators bind to secondary binding sites on the receptor and alter receptor conformation and activation, changing agonist potency and efficacy. PTC‐174 is a novel NMDA receptor positive allosteric modulator that particularly enhances activity of receptors containing GluN2C or GluN2D subunits.

Selective modulation of NaV channel gating counteracts aberrant hyperexcitability and rescues motor function and survival in a model of spinal muscular atrophy

open access: yesBritish Journal of Pharmacology, EarlyView.
Background and Purpose Spinal muscular atrophy (SMA) is a motor neuron disease caused by SMN1 gene loss, leading to reduced survival motor neuron (SMN) protein and progressive motor neuron degeneration. Although SMN‐restoring therapies improve outcomes, residual disease burden and non‐curative efficacy underscore the need for complementary treatments ...

NMDA receptor subunit‐dependent analysis of radiprodil inhibition in neonatal male rat substantia nigra dopaminergic neurons

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Background and Purpose Radiprodil is a GluN2B subunit selective NMDA receptor negative allosteric modulator which is currently under clinical investigation as a possible anti‐epileptic drug for paediatric use. Our goal was to investigate the inhibition of dopaminergic neuron NMDA responses by radiprodil.

An improved competitive inhibition enzymatic immunoassay method for tetrodotoxin quantification

open access: yesBiological Procedures Online, 2012
Quantifying tetrodotoxin (TTX) has been a challenge in both ecological and medical research due to the cost, time and training required of most quantification techniques.

Persistent in vitro nociceptor hyperexcitability and axonal retraction produced by repeated paclitaxel doses

open access: yesThe FEBS Journal, EarlyView.
Repeated paclitaxel exposure causes long‐lasting nociceptor hyperexcitability and axonal retraction in adult sensory neurons. Using a long‐term primary nociceptor culture, we show that hyperexcitability is mediated by sequential upregulation of NaV1.8, TRPV1, TRPA1, and TRPM8 channels.

Dynamic memory in rodent C‐fibres and its impact on spinal nociceptive processing

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend Nociceptive C‐fibres display activity‐dependent slowing (ADS) whereby successive action potentials are conducted progressively slower thus regulating the temporal relay of pain signals. This study shows that the ADS induced by short stimulus trains (which mimic stimulus evoked firing, shown in orange) is modified by prior ...

Tetrodotoxin Sensitivity of the Vertebrate Cardiac Na+ Current

open access: yesMarine Drugs, 2011
Evolutionary origin and physiological significance of the tetrodotoxin (TTX) resistance of the vertebrate cardiac Na+ current (INa) is still unresolved.

Trafficking and potentiation of Nav1.7 and Nav1.8 channels are mediated by IQGAP1

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend IQGAP1 binds to the voltage‐gated Nav1.7 and Nav1.8 channels and TRPA1 channels in sensory neurons. Inflammatory mediators (IM), including PGE2 and TNFα, released during nerve injury and inflammation activate intracellular signalling messengers such as PKA, p38 MAPK and Cdc42 through their respective receptors.

Spatial variability and depuration of tetrodotoxin in the bivalve Paphies australis from New Zealand

open access: yesToxicon: X, 2019
Tetrodotoxin (TTX) is a potent neurotoxin responsible for many human intoxications globally. Despite its potency and widespread occurrence in taxonomically diverse species, the primary source of TTX remains uncertain.

Tetrodotoxin and Its Analogues in Cephalothrix cf. simula (Nemertea: Palaeonemertea) from the Sea of Japan (Peter the Great Gulf): Intrabody Distribution and Secretions

open access: yesToxins, 2020
Some nemertean species from the genus Cephalothrix accumulate tetrodotoxin (TTX) in extremely high concentrations. The current study is the first to provide high-performance liquid chromatography–tandem mass spectrometry (HPLC–MS/MS) data on tetrodotoxin
