Thioacetamide toxicity in dogs has been studied. A dose of 25 mg/kg on alternate days will produce hepatic necrosis usually of the portal type, within twenty to thirty days. Six of the nine animals receiving this dose died in the stage of necrosis.

The Urease-Catalyzed Hydrolysis of Thiourea and Thioacetamide

Jack bean urease catalyzes the hydrolysis of thiourea with a second-order rate constant (kcat/Km) of 1.6 (+/- 0.2) x 10(-3) M-1 S-1 at pH7, 25 degrees C. This value is lower than that for urea by a factor of 3 x 10(8). The corresponding substitution of S for O in acetamide reduces the kcat/Km value by only a factor of 33. Abstract The mobilizing effect of thioacetomide (TAA) on deposits of mercury in the kidneys of rats was studied. Following single iv doses of mercuric chloride (HgCl 2 ) containing 0.2, 0.6, and 1.2 mg/kg of mercury, iv doses of 50 mg/kg of TAA were given 1–7 days later and continued for periods of 3–8 days.

Determination of thioacetamide by oxidation with chloramine-T

Chloramine-T oxidises thioacetamide quantitatively in an alkaline medium in the hot, converting the entire sulphur into sulphate. A back-titration method, making use of this oxidation reaction, has been suggested for the determination of thioacetamide.

Thioacetamide, which is carcinogenic in mice and rats, has been reported as negative in Ames's test on Salmonella his- with and without liver microsomal fraction (S 9 mix). Tests on Drosophila reported here showed a significant increase in sex-linked recessive lethals after treatment with thioacetamide, both after injection and after feeding of males.

Summary In thioacetamide-induced hepatomas the DNA-content decreases to diploidy without changes in the toxic swelling of the nucleus. The principle of multiplication of the nuclear volume by its (dys)functional edema also comes true in regenerative processes following partial hepatectomy.
