Thrombomodulin Structure and Function

Thrombosis and Haemostasis, 1997
Thrombomodulin is protein cofactor expressed on endothelial cell surfaces that modifies the substrate specificity of thrombin, apparently by an allosteric mechanism. The thrombin-thrombomodulin complex activates protein C, initiating an essential anticoagulant pathway.

Pharmacokinetics of recombinant human soluble thrombomodulin, thrombomodulin alfa in the rat

Xenobiotica, 2009
1. The study aimed to investigate the pharmacokinetics of thrombomodulin alpha (TM-alpha), human-soluble thrombomodulin in rats. 2. Intravenously administered TM-alpha was eliminated in two phases (T(1/2 alpha) = 0.2-0.3 h and T(1/2 beta) = 6-8 h), and the elimination curve was linear in a dose range of 10-250 microg kg(-1).

Thrombophilic thrombomodulin variants

open access: yesBlood
In this issue of Blood, Van Laer et al have expressed and extensively characterized 8 thrombomodulin (TM) variants of uncertain significance identified in patients with venous thromboembolism (VTE) or bleeding, demonstrating defective anticoagulant activity in 2 of them (L433P and C175S).1 Their findings, reported in this issue of Blood, support a ...

Loss of Thrombomodulin in Placental Dysfunction in Preeclampsia

open access: yesArteriosclerosis, Thrombosis, and Vascular Biology, 2016
OBJECTIVE: Preeclampsia is a pregnancy-specific syndrome characterized by placental dysfunction and an angiogenic imbalance. Systemically, levels of thrombomodulin, an endothelium- and syncytiotrophoblast-bound protein that regulates coagulation ...

Thrombomodulin and smoking

Atherosclerosis, 1994
Recently Blann [l] reported an increase in serum levels of von Willebrand factor antigen in smokers immediately after they had smoked two cigarettes. The von Willebrand factor is a specific endothelial constituent and its increased release indicates endothelial injury.

Thrombomodulin

Journal of Thrombosis and Haemostasis, 2003
Since its discovery as a critical cofactor in the initiation of the protein C (PC) anticoagulant pathway [1,2], biochemical and structural investigations, combined with in vivo analyses of genetically engineered mice have revealed new, and in part PC- and thrombin-independent aspects of thrombomodulin (TM) function in fibrinolysis and inflammation, and

Thrombomodulin activity and localization

The International Journal of Developmental Biology, 1993
An overview on the properties, actions and localization of thrombomodulin (TM) in situations of tissue injury and in selected tumors is presented. The localization and activity of TM after injury to vascular endothelium shows that following balloon catheter denudation of the endothelium of the rabbit aorta, the activity and immunohistochemical staining

Neuroprotection by Recombinant Thrombomodulin

Thrombosis and Haemostasis, 2000
SummaryWe examined whether recombinant human soluble thrombomodulin (rhs-TM) reduces compression trauma-induced spinal cord injury through protein C activation in rats. Administration of rhs-TM, either before or after the induction of spinal cord injury (SCI), markedly reduced the resulting motor disturbances. However, neither rhs-TM pretreated with an

The Role of Thrombomodulin in Malignancy

Seminars in Thrombosis and Hemostasis, 2007
More than 150 years ago, Trousseau first suggested a link between cancer and thrombosis. Although this link has been widely described subsequently, the underlying mechanism still remains unclear. Thrombomodulin (TM), a natural endothelial anticoagulant, has proved itself to be an exciting molecule of many functions, not least of those in inflammation ...

Thrombomodulin in sepsis.

Minerva anestesiologica, 2013
Thrombomodulin has a pivotal role in the protein C system that is important in the pathogenesis of sepsis. In sepsis, endothelial cell expression of thrombomodulin is strongly downregulated, causing an impaired activation of protein C that is central in the modulation of coagulation activation and inflammatory processes.
