Leukotriene and thromboxane antagonists

Clinical Reviews in Allergy, 1994
It has been suggested that arachidonate metabolites, leukotrienes, and thromboxane may play important roles in the pathogenesis of bronchial asthma. Biologic activities of these mediators are much more potent than those of histamine and acetylcholine on a molar basis in inducing bronchoconstriction, increase in microvascular permeability, formation of ...

A radioimmunoassay for thromboxane B2

Analytical Biochemistry, 1977
Abstract Thromboxane B 2 levels are believed to reflect the levels of the active precursor thromboxane A 2 , present during biosynthetic transformations of arachidonic acid. A radioimmunoassay for thromboxane B 2 is described. The antibodies show selective recognition of the thromboxane B structure with a sensitivity of 100 pg.

Thromboxane synthase expression and thromboxane A2 production in the atherosclerotic lesion

Journal of Molecular Medicine, 2010
Thromboxane A(2) (TXA(2)) is a potent prothrombotic and immune modulating lipid mediator, which is implicated in cardiovascular diseases, in particular, atherosclerotic lesion development and thrombogenicity. Here, we tested the hypothesis that thromboxane synthase (TXAS), the obligate enzyme required to synthesize TXA(2), is expressed within the human

Anti-thromboxane A2 actions of pinane thromboxane A2 derivatives

Prostaglandins, Leukotrienes and Medicine, 1982
Six pinane-thromboxane A2 analogs have been synthesized and tested for their ability to antagonize carbocyclic thromboxane A2 (CTA2) induced coronary vasoconstriction and prostaglandin endoperoxide analog induced platelet aggregation. Two of the derivatives, 5C-15S BPTA2 and 5T-15S BPTA2 and 5T-15S BPTA2 (1 microM) showed 76 +/- 3 and 72 +/- 9 percent ...

Glibenclamide inhibits thromboxane-mediated vasoconstriction by thromboxane receptor blockade

Vascular Pharmacology, 2004
Because sulfonylureas, such as glibenclamide, are used to treat Type 2 diabetes and because this disease is associated with various cardiovascular complications that may be mediated by thromboxane (TX), this study was designed to characterize the role of glibenclamide on TX-mediated contractions in isolated ring segments of bovine coronary arteries and

Circulating and urinary thromboxane B2 metabolites in the rabbit: 11-dehydro-thromboxane B2 as parameter of thromboxane production

Prostaglandins, 1986
The metabolism of thromboxane B2 was studied in the rabbit. The aim of the study was to identify metabolites in blood and urine that might serve as parameters for monitoring thromboxane production in vivo. [5,6,8,9,11,12,14,15-3H8]-Thromboxane B2 was administered by i.v.

CONGENITAL DEFICIENCY OF THROMBOXANE AND PROSTACYCLIN

The Lancet, 1980
Animal work suggests that with certain doses of aspirin the antithrombotic effect exerted via the inhibition of the proaggregatory platelet thromboxane A2 (TXA2) may be neutralised by the concomitant vascular reduction of the antiaggregatory prostacyclin (PGI2).

Role of Thromboxane A2

1985
When platelets are stimulated to aggregate and secrete by a sub-maximal concentration of an agonist, the platelets on the outside of an aggregate appear more degranulated than those on the inside (1) (fig. 1). In the example shown, aggregation of rabbit platelet-rich plasma (PRP) was induced by a sub-maximal concentration of collagen.

Prostaglandins and Thromboxane

Critical Care Clinics, 1985
This article attempts to delineate the role of the prostanoids in thermal injury, based on clinical and experimental trials. It also describes the authors’ concept of the cause of progressive dermal ischemia in the thermal injury and possible therapeutic measures to prevent or reduce this detrimental effect.

New Developments on Thromboxane Modulators

Mini-Reviews in Medicinal Chemistry, 2004
Thromboxane A(2) (TXA(2)) is a labile product formed from arachidonic acid by cyclooxygenase. The pathogenesis of numerous cardiovascular, pulmonary, and thromboembolic diseases can be related to this metabolite. Therefore, TXA(2) modulators have been developed for 20 years. This review will highlight the evolution in the field of TXA(2) modulators.
