Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

Numerical Analysis of the Structural Parameters on the Performance of Oil-Injected Rotary Vane Compressors

open access: yesMachines
The performance improvement potential with the optimisation of vane geometry and port timing angle in oil-injected Rotary Vane Compressors (RVCs) is not yet fully understood.

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Stable subspace dimension reduced MUSIC for blade tip timing

open access: yes, 2023
Multiple signal classification (MUSIC) is a parameter extraction method for blade tip timing to suppress the under-sampled problem. Its faster version, subspace dimension reduced MUSIC (SDR-MUSIC), has been proposed, which uses a single noise vector ...

Mass spectrometry based identification of AMP‐O‐Tris generated by Thermococcus onnurineus Cas10

open access: yesFEBS Open Bio, EarlyView.
Isolated Thermococcus onnurineus Cas10 generates the noncanonical ATP‐derived product AMP‐O‐Tris while in Tris‐containing buffer as identified via mass spectrometry, revealing relaxed nucleophile selectivity under isolated conditions. These findings suggest that multiprotein Csm complex assembly restricts Cas10 reactivity toward canonical cyclic ...

Long‐term hippocampal alterations and cognitive impairment in a murine model of surgical sepsis

open access: yesFEBS Open Bio, EarlyView.
Using a mouse model of surgical sepsis, we tested long‐term memory and analyzed the transcriptome of single cells isolated from the hippocampus. Survivor mice showed worse memory, loss of certain brain cell subpopulations, and abnormal immune cell activity—suggesting that post‐sepsis brain alterations may be linked to cognitive deficits.

A novel method for the design of proximity sensor configuration for rotor blade tip timing [PDF]

open access: yes, 2018
Blade tip timing (BTT) is a noncontact method for measuring turbomachinery blade vibration. Proximity sensors are mounted circumferentially around the turbomachine casing and used to measure the tip displacements of blades during operation.

Optimizing photoactivation of PA‐mCherry for optical pooled CRISPR screens

open access: yesFEBS Open Bio, EarlyView.
Photoactivatable PA‐mCherry finds widespread use to optically tag individual cells. However, confocal 405 nm UV laser‐scanning (normal scan) is much less efficient than widefield UV illumination, limiting the use of PA‐mCherry on confocal instruments. We remedy this limitation by reporting that rapid and repeated confocal scanning with a low‐intensity,

A BENCHMARK FOR TIP TIMING MEASUREMENT OF FORCED RESPONSE IN ROTATING BLADED DISKS [PDF]

open access: yes, 2015
The Blade Tip-Timing is a well known non-contact measurement technique for the identification of the dynamic properties of rotating bladed disks. Even if it is an industrystandard technique its reliability has to be proved for the different operation ...

Simulation of Tip-Timing Measurements of a Cracked Bladed Disk Forced Response

open access: yes, 2010
The study intends to simulate the process of the blade tip amplitude calculation by the tip-timing method. An attention is focused on tip-timing measurements for detection of a cracked blade from the bladed disk forced response.
