Anti-trypanosomal activities of DNA topoisomerase inhibitors

open access: yes, 2005
Only four drugs are available for chemotherapy of human African sleeping sickness with undesirable toxic side effects. The development of new anti-trypanosomal drugs is, therefore, urgently required.

Epigenetic Reactivation of TNFRSF19 Suppresses Mitophagy and Sensitizes Triple‐Negative Breast Cancer to Doxorubicin

open access: yesAdvanced Science, EarlyView.
TNFRSF19 is an epigenetically silenced regulator of mitophagy in triple‐negative breast cancer. TNFRSF19 deficiency activates the TGFBR1–SMAD3–PINK1 axis to promote mitophagy and confer doxorubicin resistance, whereas decitabine‐mediated restoration of TNFRSF19 suppresses mitophagy and enhances doxorubicin sensitivity, revealing a targetable epigenetic–

Plant Metabolites Screening Identifies 7‐epi‐10‐Deacetyltaxol Inhibiting Cell Division by Interfering With the Cell Cycle and Microtubule Organization

open access: yesAdvanced Science, EarlyView.
In this study, an efficient screening system is developed for plant metabolites to find a compound, 7‐epi‐10‐deacetyltaxol, inhibits tissue elongation. It suppresses plant growth through brassinosteroid and microtubule pathways. It is later applied to animal cells and human tumor organoids, resulting in similar cell division suppression. These findings

Differential in vitro activity of the DNA topoisomerase inhibitor idarubicin against Trypanosoma rangeli and Trypanosoma cruzi

open access: yesMemorias do Instituto Oswaldo Cruz, 2012
In this study the effect of eight DNA topoisomerase inhibitors on the growth Trypanosoma rangeli epimastigotes in cell culture was investigated. Among the eight compounds tested, idarubicin was the only compound that displayed promising trypanocidal ...

Activity of M3814, an Oral DNA-PK Inhibitor, In Combination with Topoisomerase II Inhibitors in Ovarian Cancer Models

open access: yesScientific Reports, 2019
DNA-dependent protein kinase (DNA-PK) has been shown to play a crucial role in repair of DNA double-strand breaks, facilitating nonhomologous end-joining. DNA-PK inhibitors have the potential to block DNA repair and therefore enhance DNA-damaging agents.

Thermal Shifts Dominate Single‐Gene Responses to Transcription‐Targeting Antibiotics

open access: yesAdvanced Science, EarlyView.
Thermal shifts reshape bacterial responses to transcription‐targeting antibiotics. In E. coli, temperature can dominate antibiotic‐induced genome‐wide expression, particularly outside 30°C–39°C, through changes in adenosine triphosphate (ATP), nucleoid organization, and drug‐target engagement.

MPP8 Facilitates DNA DSB Repair via NHEJ to Promote Chemoresistance of TNBC Independent of Methyl‐H3K9 Binding

open access: yesAdvanced Science, EarlyView.
Chemoresistance is often linked to elevated DNA repair. MPP8 is found to promote breast cancer chemoresistance as a novel NHEJ accessory factor independent of its epigenetic functions. Through multiple protein interactions, MPP8 facilitates association of DNAPK and ligase complexes to promote DNA repair progression.

Garcinol and Related Polyisoprenylated Benzophenones as Topoisomerase II Inhibitors: Biochemical and Molecular Modeling Studies.

open access: yesJournal of Natural Products, 2019
Garcinol, a polyisoprenylated benzophenone isolated from Garcinia genus, has been reported to inhibit eukaryotic topoisomerase I and topoisomerase II at concentrations comparable to that of etoposide (∼25-100 μM).
