Infant Embryonal CNS Tumors: Molecular Insights and Treatment Considerations for Contemporary Pediatric Neuro‐Oncology

open access: yesPediatric Blood &Cancer, Volume 73, Issue 11, November 2026.
ABSTRACT Background Embryonal tumors comprise the majority of malignant central nervous system (CNS) neoplasms diagnosed in children under 3 years of age. Compared with their counterparts in older children, these tumors exhibit distinct molecular biology and a more aggressive clinical phenotype, while their management is complicated by the heightened ...

Beneficial Role of Natural Polyphenols Against Epilepsy: Greasing the Wheels of Pharmacotherapy Research for Epilepsy

open access: yesFood Frontiers, Volume 7, Issue 6, November 2026.
ABSTRACT Epilepsy is the most disabling neurological disease that affects all economic classes, age groups, regions, and races. Although many antiseizure drugs are approved for epilepsy treatment, they fail to work in about 30%–40% of patients. Thus, there is a great need to identify better pharmacological agents to treat epilepsy.

Nerandomilast is Associated With Altered Alveolar Epithelial Cell State Dynamics While Attenuating Fibrotic Remodeling in Pulmonary Fibrosis

open access: yesGenes to Cells, Volume 31, Issue 6, November 2026.
Nerandomilast treatment is associated with altered AT2 cell transcriptional programs and alveolar epithelial cell state dynamics, along with attenuated fibrotic remodeling and preservation of alveolar epithelial architecture. These findings suggest that nerandomilast may modulate alveolar epithelial responses during lung injury and repair.

Diethylene Glycol‐Linked Ring‐Fused Bis‐Azoles: Target‐Oriented Synthesis, In Silico Screening, and Antitumor Activity

open access: yesChemMedChem, Volume 21, Issue 19, 14 October 2026.
A series of diethylene glycol‐linked cyclic derivatives with diversity in the heterocyclic motif have been synthesized and screened for antitumor activity. Two bis‐thiazolo[3,2‐a]pyrimidinone derivatives displayed moderate cytotoxicity, high selectivity against hepatocellular carcinoma cells, promising VEGFR2, and remarkable FGFR2 inhibitory activities

Multi‐regional Organoid Biobank Reveals FAK‐ACSL1‐Driven Doxorubicin‐Resistance and Predictive Biomarkers in Breast Cancer

open access: yesAdvanced Science, Volume 13, Issue 55, 5 October 2026.
This study established a high‐quality organoid biobank derived from 68 tumor sites across 50 Chinese patients, elucidated the drug sensitivity‐based molecular subtyping in breast cancer, and revealed a novel mechanism of drug resistance mediated by the FAK‐ACSL1 pathway.

Accelerating Primary Screening of USP8 Inhibitors from Drug Repurposing Databases with Tree‐Based Machine Learning

open access: yesAdvanced Intelligent Discovery, Volume 2, Issue 5, October 2026.
This study introduces a tree‐based machine learning approach to accelerate USP8 inhibitor discovery. The best‐performing model identified 100 high‐confidence repurposable compounds, half already approved or in clinical trials, and uncovered novel scaffolds not previously studied. These findings offer a solid foundation for rapid experimental follow‐up,

Developmental Toxicity Study for the Antimalarial Drug Combination Cabamiquine‐Pyronaridine Using a Translational Approach

open access: yesBirth Defects Research, Volume 118, Issue 10, October 2026.
ABSTRACT Background The safety of pharmacological treatments during pregnancy is a significant concern, as many medications lack comprehensive reproductive safety data. This creates gaps in therapeutic options for pregnant women, particularly malaria, which poses serious risks to maternal and fetal health.

Next‐Generation Sequencing‐Based Molecular Profiling and Management of Advanced Solid Tumors: A Real‐World Experience in a Community‐Based Academic Cancer Center

open access: yesCancer Medicine, Volume 15, Issue 10, October 2026.
ABSTRACT Background Advances in molecular and cell biology have led to a paradigm shift in cancer management. We implemented next‐generation sequencing (NGS) at our community‐based academic cancer center over a decade ago. We sought to optimize the use of genomics and understand its impact in identifying actionable genomic alterations to determine ...

Extracellularly Activatable Conjugates of RGD Peptidomimetics and Cryptophycin for αVβ3‐Targeted Cancer Therapy

open access: yesChemistry – A European Journal, Volume 32, Issue 37, 1 October 2026.
Integrin αVβ3‐targeting small‐molecule drug conjugates (SMDCs) were synthesized by conjugating a cryptophycin payload through a neutrophil elastase‐cleavable NPV‐PABC linker. RGD peptidomimetic and linker epimers served as controls for targeting and enzymatic cleavage, and the resulting conjugates displayed subnanomolar cytotoxicity in vitro.
