OUR EXPERIENCE WITH TOPOTECAN AS SECOND- LINE TREATMENT OF PATIENTS WITH RELAPSED SMALL CELL LUNG CANCER [PDF]

open access: yesJournal of IMAB, 2010
Objectives: Single agent intravenous Topotecan is an effective treatment for small cell lung cancer /SCLC/ after failure of first- line chemotherapy. The aim of this study was to evaluate the efficacy and toxicity of intravenous Topotecan in recurrent ...

Comparison of the second-line treatments for patients with small cell lung cancer sensitive to previous platinum-based chemotherapy: A systematic review and Bayesian network analysis

open access: yesFrontiers in Oncology, 2023
ObjectiveIt remains unclear what the best second-line treatment is for patients with small-cell lung cancer sensitive to previous platinum-based chemotherapy.MethodsWe systematically screened randomized controlled trials from several online databases ...

Delayed vaccine-induced CD8+ T cell expansion by topoisomerase I inhibition mediates enhanced CD70-dependent tumor eradication

open access: yesJournal for ImmunoTherapy of Cancer, 2023
Background The survival of patients with cervical cancer who are treated with cisplatin in conjunction with the topoisomerase I inhibitor topotecan is enhanced when compared with patients treated with only one of these chemotherapeutics.

Generation of the Camptothecin Scaffold by a Flavin‐Catalyzed Photooxidative Skeletal Reorganization

open access: yesAngewandte Chemie, EarlyView.
The biosynthetic pathway of the anticancer drug precursor camptothecin involves an enigmatic skeletal reorganization from a 6/5/6 tetrahydro‐β‐carboline to a 6/6/5 pyrroloquinoline ring system. Here, we show that this transformation can be achieved by photooxidation with flavin cofactors, such as flavin mononucleotide (FMN) as catalysts.

The inhibition of autophagy sensitises colon cancer cells with wild-type p53 but not mutant p53 to topotecan treatment.

open access: yesPLoS ONE, 2012
BackgroundTopotecan produces DNA damage that induces autophagy in cancer cells. In this study, sensitising topotecan to colon cancer cells with different P53 status via modulation of autophagy was examined.Methodology/principal findingsThe DNA damage ...

Structural and dynamical effects induced by the anticancer drug topotecan on the human topoisomerase I - DNA complex. [PDF]

open access: yesPLoS ONE, 2010
BACKGROUND: Human topoisomerase I catalyzes the relaxation of DNA supercoils in fundamental cell processes like transcription, replication and chromosomal segregation. It is the only target of the camptothecin family of anticancer drugs.

Lesser Toxicities of Belotecan in Patients with Small Cell Lung Cancer: A Retrospective Single-Center Study of Camptothecin Analogs

open access: yesCanadian Respiratory Journal, 2016
Purpose. Topotecan and belotecan are camptothecin derivatives that are used to treat small cell lung cancer (SCLC). This study compared the toxicities and efficacies of belotecan and topotecan monotherapies in patients with SCLC. Methods.

Inhalation delivery of topotecan is superior to intravenous exposure for suppressing lung cancer in a preclinical model

open access: yesDrug Delivery, 2018
Intravenous (IV) topotecan is approved for the treatment of various malignancies including lung cancer but its clinical use is greatly undermined by severe hematopoietic toxicity. We hypothesized that inhalation delivery of topotecan would increase local

RESILIENT Part 2: A Randomized, Open-Label Phase III Study of Liposomal Irinotecan Versus Topotecan in Adults With Relapsed Small Cell Lung Cancer

open access: yesJournal of Clinical Oncology
PURPOSE The phase III RESILIENT trial compared second-line liposomal irinotecan with topotecan in patients with small cell lung cancer (SCLC). PATIENTS AND METHODS Patients with SCLC and progression on or after first-line platinum-based chemotherapy were
