Transmissible Gastroenteritis in Neonatal Dogs: Experimental Intestinal Infection with Transmissible Gastroenteritis Virus

SUMMARY Fourteen neonatal dogs (4 through 11 days of age) were exposed orally to the Purdue strain of transmissible gastroenteritis (tge) virus, and six dogs of similar age were noninoculated controls. Clinical signs of enteric disease did not develop.

SUMMARY Fetuses in 3 sows were inoculated (intramuscularly) with transmissible gastroenteritis (tge) virus on 95th, 77th, and 74th days of the gestation. At 15, 14, and 37 days later (or days when pigs were obtained by hysterectomy), there was evidence of intestinal localization of virus, with villous atrophy and subsequent repair.

Complete Genomic Sequence of the Transmissible Gastroenteritis Virus

We have sequenced the gene 1 of TGEV Purdue-115 strain. This completes the TGEV genome sequence, which is 28579 nucleotide-long plus the poly(A) tail. TGEV is the fourth coronavirus to be completely sequenced after IBV1, MHV2,3,4 and HCV 229E5, and the second of a genetic subset which includes FIPV, CCV, HCV 229E and PEDV.

A disease with the characteristics of transmissible gastroenteritis (TGE) was first reported in 1935 (Smith, 1956). The viral etiology of TGE was demonstrated 11 years later by Doyle and Hutchings (1946) in the United States. During the next 20 years TGE was reported in all other continents (Table I).

Intracellular RNAs of an avirulent small-plaque (SP) transmissible gastroenteritis virus variant and the parent virulent Miller strain of transmissible gastroenteritis virus were compared. Northern RNA blotting showed that the Miller strain contained eight intracellular RNA species.

The Genome of Transmissible Gastroenteritis Virus (TGEV)

Lysis of isolated TGE virions with sodium dodecyl sulphate (SDS) at room temperature followed by electrophoresis, yields predominantly a single band of RNA running at about molecular weight 6.8 × 106. Heating the SDS lysate at 80°C for 15 min prior to electrophoresis results in a cluster of bands of between 0.2–6.8 × 106.
