Cancer Cell Metastasis: From Molecular Mechanisms to Therapeutic Strategies

open access: yesMedComm, Volume 7, Issue 10, October 2026.
The schematic representation of intrinsic and extrinsic influences of cancer cell metastasis. Genomic evolution of metastatic tumor cells (MTCs) promotes their metastatic phenotypes and reconstruct the primary tumor microenvironment. During MTCs migration along different routes (blood circulation, lymphatic vessels, and perineuronal spaces), the ...

TPM3–NTRK1 Fusion-Positive Thoracic Sarcoma: Integrated Diagnosis and Drug Repurposing

open access: yesIndian Journal of Medical and Paediatric Oncology
Poorly differentiated or unclassifiable sarcomas may harbor targetable molecular alterations, and next-generation sequencing is valuable for an integrated diagnosis.

Colorectal Cancer: Epidemiology, Risk Factors, Signaling Pathways, Clinical Features, Screening, Diagnosis, and Management

open access: yesMedComm, Volume 7, Issue 10, October 2026.
Colorectal cancer is a heterogeneous, adaptive disease shaped by population‐level risk, genomic and epigenetic alterations, tumor ecosystem evolution, and treatment pressure. This review integrates epidemiology, risk factors, signaling networks, clinical presentation, screening, molecular diagnosis, and stage‐ and biomarker‐guided management while ...

(R)‑2-Phenylpyrrolidine Substituted Imidazopyridazines: A New Class of Potent and Selective Pan-TRK Inhibitors

open access: yes, 2015
Deregulated kinase activities of tropomyosin receptor kinase (TRK) family members have been shown to be associated with tumorigenesis and poor prognosis in a variety of cancer types.

A De Novo Peptide That Induces Axonal Growth Through TrkB Activation

open access: yesChemBioChem, Volume 27, Issue 17, 14 September 2026.
A homodimeric anti‐TrkB macrocyclic peptide, DiTrbL3, originally discovered by the RaPID system as monomeric form, dimerizes the TrkB receptor and activate the downstream signaling. DiTrbL3 acts as a mimic of BDNF (the natural protein ligand) and stimulates hippocampal neurons to extend axons.

Molecular Characteristics of Repotrectinib That Enable Potent Inhibition of TRK Fusion Proteins and Resistant Mutations

open access: yes, 2021
NTRK chromosomal rearrangements yield oncogenic TRK fusion proteins that are sensitive to TRK inhibitors (larotrectinib and entrectinib) but often mutate, limiting the durability of response for NTRK(+) patients.

Design, Synthesis, and Evaluation of 3,5-Disubstituted 7-Azaindoles as Trk Inhibitors with Anticancer and Antiangiogenic Activities

open access: yes, 2016
Tropomyosin-related kinase A (TrkA) is considered a promising target in the development of a therapeutic treatment of cancer and pain. In this study, we designed and synthesized a series of novel 7-azaindole-based Trk kinase inhibitors through the ...

TRK Fusions Are Enriched in Cancers with Uncommon Histologies and the Absence of Canonical Driver Mutations

open access: yes, 2020
TRK inhibitors achieve marked tumor-agnostic efficacy in TRK fusion-positive cancers and consequently are now an established standard of care. Little is known, however, about the demographics, outcomes, response to alternative standard therapies, or ...

Immunological Features of Neuroendocrine Neoplasms and Adrenal Tumors

open access: yesCancer Medicine, Volume 15, Issue 9, September 2026.
ABSTRACT Neuroendocrine neoplasms, which occur throughout the human body, as well as adrenocortical carcinoma and pheochromocytoma, which originate in the adrenal gland, are primarily classified as rare malignancies. Immunotherapy, including immune checkpoint inhibitors (ICIs), is generally not incorporated into the standard care protocols for these ...
