A Universal Protein Ladder for Standardization of Diverse FRET Assays
Translating FRET measurements from in vitro to intracellular environments requires universal benchmarks. We present a modular protein ladder using TPR motifs to harmonize data across diverse platforms. This predictable calibration curve bridges single molecule FRET measurements, with cell based measurements including FLIM‐FRET.

A novel class of small RNAs: tRNA-derived RNA fragments (tRFs) [PDF]
New types of small RNAs distinct from microRNAs (miRNAs) are progressively being discovered in various organisms. In order to discover such novel small RNAs, a library of 17- to 26-base-long RNAs was created from prostate cancer cell lines and sequenced by ultra-high-throughput sequencing.

This study reveals that NOTCH2NLC transcript variant 2 generates PolyGN2C‐iso2, an aggregating protein present within intranuclear inclusions of NIID patient tissues. A novel mouse model expressing PolyGN2C‐iso2 recapitulates white matter abnormalities and cognitive deficits, mechanistically linked to mitochondrial dysfunction. These findings support a

A Panel of Circulating Exosomal sncRNAs Associated With Lung Cancer Risk up to 10 Years in Advance
Lung cancer is often diagnosed too late, and current screening overlooks many people at risk. In a long‐term study of smokers, a panel of small non‐coding RNAs carried in blood exosomes signals elevated lung cancer risk up to ten years before diagnosis, pointing toward a blood‐based tool for earlier risk detection.

A mitochondria‐derived tRNA half, mt‐5’‐tiRNA‐Tyr, is upregulated in colorectal cancer tissues and plasma and promotes tumor proliferation. It binds HARS2 and promotes CPT1A‐associated HARS2 K91 succinylation, disrupts HARS2–mt‐tRNA‐His association, impairs mitochondrial translation, enhances succinate‐linked metabolic reprogramming, and shows promise ...

A nuclear tRNA-derived fragment triggers immunity in Arabidopsis
In Arabidopsis, effector-triggered immunity (ETI) against avirulent Pseudomonas syringae pv. tomato (Pst) correlates with the rapid, Dicer-Like 1 (DCL1)-dependent nuclear accumulation of a 31-nt 5'-tRNA fragment derived from Asp-tRNA (tRF31Asp2). Several tRFs, including tRF31Asp2, are induced at early stages of infection and associate with AGO2 in the ...

Selective Export into Extracellular Vesicles and Function of tRNA Fragments during T Cell Activation
Summary: The discovery of microRNA (miRNA) sorting into extracellular vesicles (EVs) revealed a novel mode of intercellular communication and uncovered a link between cellular endomembrane compartments and small RNAs in EV-secreting cells.

Objective Microbial small RNAs can regulate human genes. Higher plasma concentrations of microbial transfer RNA (tRNA)‐derived RNA‐1 (tDR‐1) were previously associated with lower rheumatoid arthritis (RA) disease activity. This study examined whether tDR‐1 concentrations differ in anticyclic citrullinated peptide‐3 positive (CCP3+) at‐risk individuals (

ABSTRACT Neisseria gonorrhoeae poses an urgent public health challenge due to rapidly increasing antimicrobial resistance and the absence of an effective vaccine. Targeting conserved bacterial pathways involved in essential physiological processes may provide new opportunities for vaccine antigen discovery.
