Epigenetic Silencing of RFX7 Defines a Transcriptional Axis Linking Lactate Metabolism to Immune Checkpoint Therapy in Glioblastoma

open access: yesAdvanced Science, EarlyView.
Glioblastoma (GBM) exhibits RFX7 silencing via promoter hypermethylation, which downregulates PIK3IP1 to activate PI3K/AKT signaling, increase lactate accumulation, and enhance histone H4K12 lactylation. This lactylation upregulates PD‐L1/CSF1 expression and drives the immunosuppressive microenvironment formation.

Current Concepts of Tumor-Infiltrating Lymphocytes in Human Malignancies

open access: yes, 2009
Tumor-infiltrating lymphocytes (TILs) develop as manifestations of the recognition and defense against malignant cells by the host immune system. TILs were literally defined as “tumor-infiltrating lymphocytes”,which a posteriori locate within the tumor ...

Tumor‐Intrinsic ARHGEF3 Enhances Antitumor Immunity by Promoting T‐Cell Infiltration and Limiting Myeloid Cell‐Mediated Immunosuppression

open access: yesAdvanced Science, EarlyView.
ARHGEF3 is broadly downregulated across human cancers and correlates with patient prognosis. Tumor‐intrinsic ARHGEF3 activates the RHOA–ROCK–PTEN cascade to inhibit AKT signaling, thereby promoting chemokine‐driven T‐cell infiltration and relieving lipid‐mediated myeloid immunosuppression.

Bioorthogonal Fluorogenic Reporters for Noninvasive Imaging and Urinalysis of Immunotherapeutic Response in Renal Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
This study reported renal‐clearable bio‐orthogonal near‐infrared fluorogenic probes (BGRs) that specifically imaging and urinalysis of granzyme B for dynamic evaluation of RCC immunotherapy. BGRs not only differentiate immunotherapeutic responses in orthotopic RCC mice, but also enable sensitive optical urinalysis of granzyme B in clinical specimens ...

Tumor‐Immune‐On‐Chip to Evaluate Pathophysiological Feature of T Lymphocytes Expanded from Patient Tumors and Lymph Node Tissues

open access: yesCancer Medicine
The infiltration and cytotoxicity of T lymphocytes are critical for cancer immunotherapy efficacy; however, the behavior of these immune cells has not been thoroughly investigated.

A Hydrogen‐Releasing Nanozyme Engineers a Mitochondrial ROS Amplifier for Self‐Sustaining Catalytic Immunotherapy

open access: yesAdvanced Science, EarlyView.
We developed a RhPd‐H nanozyme that integrates enhanced POD‐mimetic catalytic activity with thermally triggered H2 release. H2 shields and reprograms mitochondria to continuously leak endogenous ·O2−. This dual‐path ROS spatially and temporally disrupts redox homeostasis, inducing persistent oxidative stress for tumor suppression and antitumor immunity,

Ten-year clinical outcome, toxicity and compliance of dose-dense sequential adjuvant administration of cyclophosphamide & epirubicin followed by docetaxel in patients with early breast cancer: A hellenic cooperative oncology group observational study (HE 10/10) with concurrent investigation of significance of tumor infiltrating lymphocytes

open access: yesBreast
Background: Dose-dense sequential (dds) chemotherapy has changed the clinical outcome of patients with early breast cancer (BC). To investigate the impact of dose intensity (DI) in the adjuvant setting of BC, this observational trial (HE 10/10) was ...
