H3K36me3‐Guided m6A Modification of Oncogenic L1CAM‐AS1 Drives Macrophage Polarization and Immunotherapy Resistance in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
L1CAM‐AS1 is identified as a novel H3K36me3‐guided, m6A‐modified long noncoding RNA (lncRNA) in hepatocellular carcinoma (HCC) cells. L1CAM‐AS1 stabilizes RAN protein, enhances M2 macrophages‐released CCL5‐induced nuclear import of RELA, activates the NF‐κB signaling, up‐regulates CCL2 expression and secretion from HCC cells, and enhances M2 ...

The Balance between Mono- and NEDD8-Chains Controlled by NEDP1 upon DNA Damage Is a Regulatory Module of the HSP70 ATPase Activity

open access: yesCell Reports, 2019
Summary: Ubiquitin and ubiquitin-like chains are finely balanced by conjugating and de-conjugating enzymes. Alterations in this balance trigger the response to stress conditions and are often observed in pathologies.

An acidic loop and cognate phosphorylation sites define a molecular switch that modulates ubiquitin charging activity in Cdc34-like enzymes. [PDF]

open access: yesPLoS Computational Biology, 2011
E2 ubiquitin-conjugating enzymes are crucial mediators of protein ubiquitination, which strongly influence the ultimate fate of the target substrates.

Interactions between the quality control ubiquitin ligase CHIP and ubiquitin conjugating enzymes

open access: yesBMC Structural Biology, 2008
BackgroundUbiquitin (E3) ligases interact with specific ubiquitin conjugating (E2) enzymes to ubiquitinate particular substrate proteins. As the combination of E2 and E3 dictates the type and biological consequence of ubiquitination, it is important to ...

The Ubc3 (Cdc34) Ubiquitin-Conjugating Enzyme Is Ubiquitinated and Phosphorylated In Vivo [PDF]

open access: yesMolecular and Cellular Biology, 1994
The transition from G1 to S phase of the cell cycle in Saccharomyces cerevisiae requires the activity of the Ubc3 (Cdc34) ubiquitin-conjugating enzyme. S. cerevisiae cells lacking a functional UBC3 (CDC34) gene are able to execute the Start function that initiates the cell cycle but fail to form a mitotic spindle or enter S phase.

Listerin Alleviates Alzheimer's Disease through IRE1‐mediated Decay of TLR4 mRNA

open access: yesAdvanced Science, EarlyView.
Alzheimer's disease (AD) progression is influenced by microglia‐mediated neuroinflammation. Here, it is demonstrated that Listerin suppresses neuroinflammatory signaling and cognitive impairment in AD models by triggering IRE1α‐mediated TLR4 mRNA decay. Adenoviral Listerin delivery reduces amyloid‐β pathology, positioning it as a new therapeutic target.

E3 ubiquitin ligases: styles, structures and functions

open access: yesMolecular Biomedicine, 2021
E3 ubiquitin ligases are a large family of enzymes that join in a three-enzyme ubiquitination cascade together with ubiquitin activating enzyme E1 and ubiquitin conjugating enzyme E2.

Two different classes of E2 ubiquitin-conjugating enzymes are required for the mono-ubiquitination of proteins and elongation by polyubiquitin chains with a specific topology.

open access: yesBiochemical Journal, 2008
RING (really interesting new gene) and U-box E3 ligases bridge E2 ubiquitin-conjugating enzymes and substrates to enable the transfer of ubiquitin to a lysine residue on the substrate or to one of the seven lysine residues of ubiquitin for polyubiquitin ...

Natural Allelic Variations in IbCHYR1–IbZnFR Complex Regulate Fusarium Root Rot Resistance in Sweet Potato

open access: yesAdvanced Science, EarlyView.
FfRlpA2, a conserved Fusarium effector, functions as a protease inhibitor by hijacking the E3 ubiquitin ligase IbCHYR1 to degrade the resistance factor IbZnFR, thereby promoting Fusarium pathogenicity. Conversely, the high dosage Pro::IbZnFRHap2 allele was associated with resistance to root rot disease.
