Preservation of ALYREF Phase Separation Mitigates Doxorubicin‐Induced Cardiomyocyte DNA Damage and Cardiotoxicity

open access: yesAdvanced Science, EarlyView.
Binding of Doxorubicin to ALYREF disrupts its phase‐separated condensate and induces DNA damage and apoptosis in cardiomyocytes Abstract The clinical utility of the anticancer agent doxorubicin (DOX) is limited by its dose‐dependent cardiotoxicity.

Targeting Methylglyoxal Metabolism to Enhance Ferroptosis Sensitivity in Tumor Therapy

open access: yesAdvanced Science, EarlyView.
This study reveals how blocking a detox enzyme called GLO1 increases the buildup of a toxic molecule, methylglyoxal (MGO), which triggers cancer cell death by ferroptosis. A protein called PJA1 drives this process. Targeting the PJA1–GLO1 pathway boosts the effectiveness of ferroptosis‐inducing treatments in tumors, offering a promising approach for ...

Discovering Uncharted Binding Pockets on E3 Ligases Leads to the Identification of FBW7 Allosteric Modulators

open access: yesAdvanced Science, EarlyView.
A computational approach to identify and characterize ligandable binding pockets on E3 ligases is presented. As a validation case, the first low‐micromolar small moleculesthat bind to the FBW7 E3 ligase are identified and confirmed to bind at the intended site.

Disruption of NF‐κB‐Mediated Copper Homeostasis Sensitizes Breast Cancer to Cuproptosis

open access: yesAdvanced Science, EarlyView.
This study reveals a feedback mechanism regulating copper homeostasis, in which copper directly interacts with TAK1 to activate NF‐κB signaling, while NF‐κB transcriptionally suppresses copper transporter (CTR1) expression. These findings highlight a promising therapeutic strategy for breast cancer by combining NF‐κB inhibitors with copper chelators or

Post‐Translational Modifications of TOE3 Regulate Antiviral Defense in Tobacco

open access: yesAdvanced Science, EarlyView.
CK2αL phosphorylates TOE3 and enhances its stability, while FBXL1 promotes the ubiquitination‐degradation of TOE3 via the 26S proteasome. Compared to CK2αL‐phosphorylated TOE3, nonphosphorylated TOE3 shows a much higher affinity for FBXL1 and is more susceptible to ubiquitination‐degradation, thus facilitating viral infection. The regulatory mechanisms
