Impact of Obesity and MASH on Zonal Hepatocellular Statin Exposure: Pharmacodynamic Insights From a Permeability‐Limited Multicompartment Liver Model

open access: yesCPT: Pharmacometrics &Systems Pharmacology, Volume 15, Issue 1, January 2026.
Obesity, MASH and Statins: Insights from a PBPK/PD Zonal Liver Model. ABSTRACT Statins are frequently prescribed for hyperlipidemia, a common comorbidity in patients with obesity and/or metabolic dysfunction‐associated steatohepatitis (MASH). However, limited knowledge exists on how MASH may alter statin disposition within hepatocytes where the statin ...

Physiologically‐Based Pharmacokinetic Modeling to Support Pediatric Clinical Development: An IQ Working Group Perspective on the Current Status and Challenges

open access: yesCPT: Pharmacometrics &Systems Pharmacology, Volume 15, Issue 1, January 2026.
ABSTRACT Pediatric extrapolation strategies issued by health authorities have streamlined pediatric drug development and reduced the unnecessary burden of conducting pediatric clinical studies. In line with these strategies, physiologically based pharmacokinetic (PBPK) models have been utilized extensively for initial dosing regimen and sampling ...

Triplet RNA Lipid Nanoparticles for Locoregional Cancer Immunotherapy

open access: yesSmall Science, Volume 6, Issue 1, January 2026.
LNPs deliver the immunoadjuvant pIpC into tumors to activate local cells. After activation, additional RNA (siRNA or mRNA) can modulate immune pathways by suppressing immunoinhibitory targets or inducing immunostimulatory ones. This coordinated “triplet RNA” approach may enhance immune activation within the tumor microenvironment.

Nanozyme‐Integrated 3D‐Printed Gradient Scaffold Rescues Redox Homeostasis for Enhanced Osteochondral Repair

open access: yesSmall Structures, Volume 7, Issue 1, January 2026.
This work presents a novel 3D‐printed gradient scaffold for osteochondral repair. The implant features distinct layers, each designed for a specific therapeutic role: a cartilage layer protects tissue and a bone layer promotes regeneration. In vivo experiments confirm the scaffold's effectiveness, demonstrating its dual ability to maintain chondrocyte ...
