We have encapsulated a host–guest reporter pair together with three enzymes within vesicles to monitor a two‐compartment cascade reaction which includes transmembrane transport of peptide substrates and their subsequent enzymatic conversion inside the vesicles. Our “protocell” system mimics a simplistic cellular uptake‐metabolism cycle.
