The ''Rainbow western'' method permits detection of multiple antigens on a single protein blot. The procedure utilizes horseradish peroxidase (HRPase)-based detection with both a chemiluminescent and colorimetric substrate. In the ''Rainbow western'' procedure four different HRPase-colorimetric substrates that produce black, brown, red, and green ...

Protein-protein interactions play an important role in the proper functioning of the cell. Far-western blotting is a useful technique to study protein-protein interactions. In this method, proteins of interest are attached to a membrane and then probed with a non-antibody protein.

Immunodetection of Proteins by Western Blotting
2003The technique of protein immunoblotting, more commonly known as "Western" blotting, was first described a decade ago by Towbin et al., (1), using electric current to transfer polypeptides from polyacrylamide gels to nitrocellulose, although several groups had previously reported the use of capillary forces to effect the transfer of DNA ("Southern ...

Coomassie staining as loading control in Western blot analysis.
Journal of Proteome Research, 2011In Western blotting, immunodetection of housekeeping proteins is routinely performed to detect differences in electrophoresis loading. The present work describes a much faster and simpler protein staining method, which is compatible with ordinary ...

Antibody Validation by Western Blotting
2017Validation of antibodies is an integral part of translational research, particularly for biomarker discovery. Assaying the specificity of the reagent (antibody) and confirming the identity of the protein biomarker is of critical importance prior to implementing any biomarker in clinical studies, and the lack of such quality control tests may result in ...

Every cell contains several proteins. How do we identify every protein in the cell? This question vexed many scientists until the introduction of the procedure known as protein blotting or Western blotting. Cells obtained from tissues or cells grown outside the human body in a plastic flask are first busted to release its contents.

This chapter provides an overview of the western blot analysis method, which allows the investigator to identify specific proteins resolved by SDS polyacrylamide gel electrophoresis by binding with specific antisera. Proteins resolved on an acrylamide gel are transferred to a nitrocellulose (NC) filter that is incubated with the antisera.

Western Blotting Using Microfluidics
2015Together with polyacrylamide gel electrophoresis, the western blot has been an invaluable research technique in biological sciences. It continues to serve as an important diagnostic tool in medical laboratories. The procedure, however, involves multiple steps that are often time and resource intensive in addition to being of low throughput.
