Unraveling Suicidal Ideation and Behaviors in a Global Cohort of Individuals at Clinical High‐Risk for Psychosis: Multivariable Correlates and Subthreshold Psychotic Symptoms

open access: yesActa Psychiatrica Scandinavica, EarlyView.
ABSTRACT Introduction Suicidal ideation and behaviors (SIB) are common among individuals at clinical high risk for psychosis (CHR‐P), but their relationship with the overall clinical profile remains unclear. This study aimed to identify sociodemographic/clinical factors associated with SIB in CHR‐P individuals and to examine whether subthreshold ...

Lung function response to 12-week treatment with combined inhalation of long-acting β2 agonist and glucocorticoid according to ADRB2 polymorphism in patients with chronic obstructive pulmonary disease

open access: yes, 2018
Recent reports suggest that β2-adrenergic receptor (ADRB2) genotypes are associated with therapeutic responses to β2 agonists in asthmatics. However, few studies have investigated therapeutic responses to β2 agonists in chronic obstructive pulmonary ...

Deficiency of Mitochondrial Fatty Acid Enzyme, CPT1A, Underlies Airway Epithelial Barrier Dysfunction in Severe Asthma

open access: yesAllergy, EarlyView.
Mitochondrial fatty acid enzyme, CPT1A, is deficient in the airway epithelium of severe asthma patients. Restoration of CPT1A expression improved epithelial barrier integrity via increasing mitochondrial respiration and ATP production in airway epithelial cells. Restoration of deficient epithelial CPT1A may represent a new treatment approach for severe

Scalable Ruthenium-Catalyzed Asymmetric Synthesis of a Key Intermediate for the β2-Adrenergic Receptor Agonist

open access: yes, 2016
An enantioselective and robust synthetic process to obtain a useful intermediate for the β2-adrenergic receptor agonist is described. Asymmetric transfer hydrogenation of ketone 1c by (S,S)-3b (Ms-DENEB) afforded chiral alcohol 2c in 71% isolated yield ...

Pathogenic Airway Bacteria and Local Immune Responses During Early‐Life Asthma‐Like Episodes

open access: yesAllergy, EarlyView.
This study investigates how Haemophilus influenzae, Streptococcus pneumoniae, and Moraxella catarrhalis associate with airway immune responses during early‐life asthma‐like episodes, and compare these responses between children with and without asthma.

Rademikibart for Asthma With Type 2 Biomarkers: Exploratory Subgroup Analyses From a Phase 2b Randomized Clinical Trial

open access: yesAllergy, EarlyView.
This study reports exploratory and predominantly post hoc subgroup analyses, according to baseline eosinophil count and FeNO levels. In adults with moderate‐to‐severe uncontrolled asthma, rademikibart produced rapid and sustained improvements in lung function and asthma control.

Compartmentalisation in cAMP signalling: A phase separation perspective

open access: yesBritish Journal of Pharmacology, EarlyView.
Cells rely on precise spatiotemporal control of signalling pathways to ensure functional specificity. The compartmentalisation of cyclic AMP (cAMP) and protein kinase A (PKA) signalling enables distinct cellular responses within a crowded cytoplasmic space.

The potential for biased signalling in the P2Y receptor family of GPCRs

open access: yesBritish Journal of Pharmacology, EarlyView.
The purinergic receptor family is primarily activated by nucleotides, and contains members of both the G protein coupled‐receptor (GPCR) superfamily (P1 and P2Y) and ligand‐gated ion channels (P2X). The P2Y receptors are widely expressed in the human body, and given the ubiquitous nature of nucleotides, purinergic signalling is involved with a plethora

The discovery and development of ensifentrine: A novel inhaled dual PDE3/4 inhibitor having ‘bifunctional’ bronchodilator and anti‐inflammatory activity

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Ensifentrine (formerly called RPL 554) is a novel, first in class, inhaled bifunctional dual PDE3/4 inhibitor for the treatment of chronic respiratory diseases. Ensifentrine exhibits both bronchodilation via PDE3 inhibition and anti‐inflammatory activity via PDE4 inhibition.
