外源基因转染对大肠癌细胞及放射敏感性的影响

目的探讨p16基因转染对大肠癌细胞生长及对放射敏感性的作用。方法以脂质体介导外源性P16基因转染大肠癌SWWC细胞系,照射后MTT法测定OD值,计算细胞生长抑制率。结果p16基因转染大肠癌SWWC细胞发生周期阻滞,照射后转染组细胞生长抑制率增加,与对照组有明显差异。结论p16基因转染对大肠癌SWWC细胞具有明显抑制作用,可提高大肠癌细胞对放射的敏感性。

肺癌是常见肺原发恶性肿瘤,近年来随着EGFR抑制剂等基因靶向药物的问世,肺癌的治疗取得了长足的进步。吉非替尼是EGFR-TKI类靶向治疗的代表性药物,是一种口服的选择性EGFR酪氨酸激酶抑制剂。RBM5作为一个肿瘤抑制基因,前期研究发现RBM5与肺癌化疗耐药有关,可逆转肺腺癌细胞顺铂耐药性。本研究通过体外实验,检测RBM5过表达对吉非替尼耐药细胞EGFR基因表达及细胞增殖的影响,探讨RBM5在改善吉非替尼耐药中的作用,探寻其逆转耐药机制。
