重组腺病毒介导RECK基因在大肠癌细胞中的表达及抑制癌细胞浸润能力的实验研究

目的 研究重组腺病毒介导RECK基因转染大肠癌细胞后基因的表达情况,并观察转染细胞浸润能力变化。方法 构建了RECK基因的复制缺陷型重组腺病毒载体adRECK ,以adRECK转染大肠癌癌细胞株后,用RT PCR法检测RECK基因在大肠癌中的表达。并用Boyden小室测试转染细胞的侵袭性。结果 RECK基因在转染细胞中有效的表达。被转染细胞浸润能力明显下降。结论 重组腺病毒介导RECK基因能够在大肠癌细胞中有效的表达,转染病毒的大肠癌浸润能力明显下降。
