ARC3 levels are controlled by cytosolic and chloroplast proteolytic systems. PUB52 mediates ARC3 precursor ubiquitination and degradation in the cytosol, while CLPC1 promotes ARC3 degradation in chloroplasts, where ARC2 protects ARC3 from excessive breakdown. Disrupting these components causes chloroplast division defects, placing them upstream of ARC3.

Dysregulated protein modifications drive tumorigenesis. RINES, an E3 ubiquitin ligase, represses tumor cell proliferation and metastasis by facilitating RING domain‐dependent, ubiquitin–proteasome‐mediated degradation of STAT3 and MYC, which consequently restrains cancer stemness and oncogenic progression.

ABSTRACT Astrocyte reactivity critically shapes neuroinflammatory outcomes after ischemic stroke, yet the upstream regulators governing astrocyte state transitions remain incompletely defined. Here, we identify the immunoproteasome subunit low molecular weight protein 2 (LMP2) as an important modulator of astrocyte functional remodeling following ...

Non-Covalent Inhibitors of the 20S Proteasome
Peptidomimetics have been commonly used as lead compounds to design inhibitors with high affinity and specificity for a particular enzyme. The discovery that a 2-aminobenzylstatine derivative originally designed to target an aspartyl protease ...

Mitochondrial SLC25A46 is upregulated in ovarian cancer (OC) and correlates with poor prognosis. SLC25A46 stabilizes CACT to activate fatty acid oxidation, increasing ATP and NADPH production to drive cell proliferation and ferroptosis evasion. SLC25A46 silencing sensitizes OC cells to ferroptosis and carboplatin, offering a promising therapeutic ...

Early cysteine-dependent inactivation of 26S proteasomes does not involve particle disassembly
Under oxidative stress 26S proteasomes suffer reversible disassembly into its 20S and 19S subunits, a process mediated by HSP70. This inhibits the degradation of polyubiquitinated proteins by the 26S proteasome and allows the degradation of oxidized ...

The 20S Proteasome Splicing Activity Discovered by SpliceMet
The identification of proteasome-generated spliced peptides (PSP) revealed a new unpredicted activity of the major cellular protease. However, so far characterization of PSP was entirely dependent on the availability of patient-derived cytotoxic CD8+ T lymphocytes (CTL) thus preventing a systematic investigation of proteasome-catalyzed peptide splicing

ABSTRACT Iron overload is a common metabolic disturbance in cancer and contributes to poor outcomes in renal cell carcinoma (RCC), yet its effects on the tumour immune microenvironment remain unclear. Here we identify a previously unrecognized immunosuppressive axis in which iron overload downregulates the palmitoyltransferase ZDHHC12 in CD8+ T cells ...

Decelerated degradation of short peptides by the 20S proteasome
Based on a twelve residue master peptide comprising all five specific cleavage sites defined for the proteasome, a set of variant peptides was generated in order to probe specificity and to elucidate the mechanism which determines product size. It is shown that the rate of degradation by the 20S proteasome from Thermoplasma acidophilum depends ...

NLSS3 Impairs SHM1 Autophagic Degradation to Regulate Leaf Morphology and Salt Tolerance in Rice
In rice, NLSS3 binds and shields SHM1 from autophagic degradation to maintain serine homeostasis. The A132P mutation in nlss3 disrupts this, triggering SHM1 loss, metabolic dysfunction, K+ imbalance, and impaired ROS scavenging, which collectively drive narrow leaves and salt sensitivity.
