Effect of 1α,25‐dihydroxyvitamin D3 in embryonic hippocampal cells [PDF]

open access: possibleHippocampus, 2009
AbstractAlthough the role of 1α,25‐dihydroxyvitamin D3 in calcium homeostasis of bone tissue is clear, evidence of the involvement of vitamin D3 in the central nervous system functions is increasing. In fact, vitamin D3 regulates vitamin D receptor and nerve growth factor expression, modulates brain development, and reverses experimental autoimmune ...

24R,25-dihydroxyvitamin D3 and 1α,25-dihydroxyvitamin D3 are both indispensable for calcium and phosphorus homeostasis

Life Sciences, 1980
Abstract The essential role of vitamin D throughout the life of most mammals and birds as a mediator of calcium homeostasis is well established. In view of the complex endocrine system existent for the regulated metabolism of vitamin D 3 to both 1α,25-dihydroxyvitamin D 3 [1,25(OH) 2 D 3 ] and 24R,25-dihydroxyvitamin D 3 [24R,25-(OH) 2 D 3 ] (both

Chemical synthesis of [1β-3H]1α,25-dihydroxyvitamin D3 and [1α-3H]1β,25-dihydroxyvitamin D3: Biological activity of 1β,25-dihydroxyvitamin D3

Biochemical and Biophysical Research Communications, 1980
The simple three-step preparation of [1β-3H]1α,25-dihydroxyvitamin D3 and [1α-3H]1β,25-dihydroxyvitamin D3 from 1α,25-dihydroxyvitamin D3 is described. In the rat, 1β,25-dihydroxyvitamin D3, when compared with its α-epimer, did not stimulate intestinal calcium transport or bone calcium mobilization at doses 1000-fold higher than the doses of the ...

The Effects of 24R, 25-dihydroxyvitamin D3 and 24S, 25-dihydroxyvitamin D3 on Phosphate Transport in Chicks In Vivo

Immunology‚ Endocrine & Metabolic Agents in Medicinal Chemistry, 2013
Previous work in this laboratory has indicated that 24,25-dihydroxyvitamin D3 (24,25(OH)2D3) inhibits rapid, 1,25-dihuydroxyvitamin D3 (1,25(OH)2D3) stimulated phosphate uptake in isolated intestinal cells and perfused duodenal loops. Studies were undertaken to determine if 24,25(OH)2D3 had a similar effect in vivo.

Stimulation of 24R,25-dihydroxyvitamin D3 synthesis by metabolites of vitamin D3

American Journal of Physiology-Endocrinology and Metabolism, 1983
Previously we have shown that the isolated perfused kidney from vitamin D-deficient rats converts [3H]25(OH)D3 into [3H]1 alpha,25(OH)2D3. When certain vitamin D metabolites were added to perfusate the same kidney began to synthesize [3H]24R,25(OH)2D3.

A short practical approach to 24R,25-dihydroxyvitamin D3

The Journal of Steroid Biochemistry and Molecular Biology, 2010
A synthesis of the vitamin D3 metabolite 24R,25-dihydroxyvitamin D3 (1) by Lythgoe's Wittig-Horner approach is described. The key step of the synthesis is the stereocontrolled introduction of the 24-hydroxyl group by a palladium(0)-induced [3,3]-sigmatropic rearrangement on a 22R-allylic acetate (7).

Specific binding of 24R,25-dihydroxyvitamin D3 to chick intestinal mucosa: 24R,25-dihydroxyvitamin D3 is an allosteric effector of 1,25-dihydroxyvitamin D3 binding

Archives of Biochemistry and Biophysics, 1986
We have previously described a significant decrease in the positive cooperativity level and affinity of 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] binding to its chick intestinal chromatin receptor induced in vitro by a physiological 10-fold molar excess of (24R)-25-dihydroxyvitamin D3 [24R,25(OH)2D3] [F. The site of 1α,25-dihydroxyvitamin D3 production in pregnancy

Biochemical and Biophysical Research Communications, 1979
Metabolism of 25-hydroxyvitamin D3 (25-OH-D3) in pregnancy was investigated invitro in New Zealand White rabbits fed a rabbit chow. Kidney homogenates from pregnant mothers and fetuses were separately incubated with [3H]-25-OH-D3. The homogenates from fetuses produced significant amounts of [3H]-1α,25-dihydroxyvitamin D3 [1α,25-(OH)2-D3] from its ...

Effects of the vitamin D3 analog 1α,25-dihydroxyvitamin D3-3β-bromoacetate on rat osteosarcoma cells: Comparison with 1α,25-dihydroxyvitamin D3

Journal of Cellular Biochemistry, 1996
The actions of the hormonal form of vitamin D, 1 alpha, 25-dihydroxyvitamin D3 [1 alpha, 25-(OH)2 D3], are mediated by both genomic and nongenomic mechanisms. Several vitamin D synthetic analogs have been developed in order to identify and characterize the site(s) of action of 1 alpha, 25-(OH)2D3 in many cell types including osteoblastic cells. We have

Fluorometric assay of 25-hydroxyvitamin D3 and 24R,25-dihydroxyvitamin D3 in plasma

Analytical Biochemistry, 1992
The first practical fluorometric assay of plasma 25-hydroxyvitamin D3 (25-OH-D3) and 24R,25-dihydroxyvitamin D3 (24,25-(OH)2D3) is described. The method uses a highly fluorescent dienophile, 4-[2-(6,7-dimethoxy-4-methyl-3-oxo-3,4-dihydroquinoxalyl)ethyl]-1, 2,4- triazoline-3,5-dione (DMEQ-TAD), to fluorescence-label vitamin D.
