An Injectable Metal‐Drug Supramolecular Condensate Enables Radiosensitization and Systemic Antitumor Immunity for Post‐Surgical Osteosarcoma Therapy

open access: yesAdvanced Science, EarlyView.
This work designed an injectable metal‐drug supramolecular condensate formed from hafnium ions and alendronate (Hf‐ALN) and loaded with the COX‐2 inhibitor celecoxib (CXB) for synergistic radioimmunotherapy of postoperative osteosarcoma. Mechanistically, upon X‐ray irradiation, Hf enhances radiation energy deposition, ROS generation, and DNA damage ...

Experimental model for the irradiation-mediated abscopal effect and factors influencing this effect [PDF]

open access: yes, 2020
Radiotherapy (RT) is the primary treatment for cancer. Ionizing radiation from RT induces tumor damage at the irradiated site, and, although clinically infrequent, may cause regression of tumors distant from the irradiated site-a phenomenon known as the ...

Glutaminolysis Blockade‐Empowered Bimodal Nanodepot for Spatially Complementary Sono‐Thermal Ablation via PANoptosis and STING Activation Against Large Tumors

open access: yesAdvanced Science, EarlyView.
An injectable bimodal TCPD nanodepot disrupts glutaminolysis and TCA metabolism, sensitizing large tumors to spatially complementary sono‐thermal ablation for complete treatment coverage. This intervention induces mitochondrial and endoplasmic reticulum dysfunction, genotoxicity, and NAD+ depletion, converging to trigger PANoptosome assembly and ...

The abscopal effects of sonodynamic therapy in cancer

open access: yesBritish Journal of Cancer
Abstract The abscopal effect is a phenomenon wherein localised therapy on the primary tumour leads to regression of distal metastatic growths. Interestingly, various pre-clinical studies utilising sonodynamic therapy (SDT) have reported significant abscopal effects, however, the mechanism remains largely enigmatic.

Targeting Macrophage‐Mediated Lymphatic Immunosuppression to Prevent Tumor Recurrence After Microwave Ablation

open access: yesAdvanced Science, EarlyView.
Incomplete microwave ablation induces animmunosuppressive microenvironment in tumor‐draining lymph nodes. An albumin‐based lipoprotein nanosystem enables coordinated delivery of immunomodulators to tumor‐draining lymph nodes and residual tumors, reverses post‐ablation immunosuppression, suppresses residual tumor growth, and promotes durable antitumor ...

Immune mechanisms mediating abscopal effects in radioimmunotherapy

open access: yesPharmacology & Therapeutics, 2019
Radiotherapy of cancer has been traditionally considered as a local therapy without noticeable effects outside the irradiated fields. However, ionizing radiation exerts multiple biological effects on both malignant and stromal cells that account for a complex spectrum of mechanisms beyond simple termination of cancer cells. In the era of immunotherapy,

Bioengineered Stroma‐Camouflaged Nanovesicles Disrupt the IDO1/CD276 Immunosuppressive Circuit to Reinvigorate Antitumor Immunity

open access: yesAdvanced Science, EarlyView.
Bioengineered stroma‐camouflaged nanovesicles (AM@UIO‐BMS/DOX) convert the immunosuppressive tumor niche into an immunoresponsive milieu by disrupting the IDO1/CD276 circuit. Anti‐CD276 membrane camouflage guides tumor accumulation and deep penetration, enabling controlled co‐delivery of DOX and the IDO1 inhibitor BMS‐986205.

Abscopal Effect of Radiation Therapy and Signal Transduction

open access: yesCurrent Signal Transduction Therapy, 2010
The abscopal effect is a potentially important phenomenon as a topic for basic research on tumor control and clinical oncology. Although it has been described in various malignancies, it is a rarely recognized clinical event. This phenomenon may arise mainly as a result of an activated immune system mediated through cytokines.

Low‐Density Lipoprotein Receptor‐Targeted Biomimetic Nanoplatform Suppresses Glioblastoma Stemness and Remodels Antitumor Immunity via Dual Metabolic Disruption

open access: yesAdvanced Science, EarlyView.
A low‐density lipoprotein receptor‐oriented biomimetic nanoplatform couples brain‐tumor delivery with a mechanistically matched regorafenib‐erythrodiol combination. Concurrent disruption of autophagic nutrient salvage and cholesterol homeostasis collapses bioenergetic and signaling support, suppresses glioblastoma stemness, and promotes immunogenic ...
