5‐HT2A receptor agonism by tert‐leucinamide and valinamide synthetic cannabinoids: In vitro and in vivo evidence

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Background and Purpose New synthetic cannabinoid receptor agonists (SCRAs) are associated with severe adverse effects, including unexpected psychiatric symptoms. These compounds are mainly active through their potent agonism on the cannabinoid receptors CB1 and CB2.

Conversion of thioacetamides, acetamides, and acetonitriles into trimethinium salts under Vilsmeier conditions

open access: yes, 1987
Thioacetamides and N-(3-aminothioacryloyl)formamidines can be transformed to trimethinium salts by the reaction with formamide chlorides. The same products are formed by a modified Vilsmeier-Haack-Arnold reaction of acetamides or acetonitriles.

Understanding how enzymes work: the journey to ensemble–function studies

open access: yesThe FEBS Journal, EarlyView.
For decades, structure–function has dominated biochemistry. Structures are highly valuable, yet more is needed to achieve a quantitative understanding of biomolecular function, because function emerges from an ensemble of states, rather than a static structure. We describe an ensemble–function framework applied to quantitatively dissect serine protease

Organic synthesis using (diacetoxyiodo)benzene (DIB): Unexpected and novel oxidation of 3-oxo-butanamides to 2,2-dihalo-N-phenylacetamides

open access: yesBeilstein Journal of Organic Chemistry, 2012
A novel and reliable method for the direct preparation of 2,2-dihalo-N-phenylacetamides is reported. The key transformation involves the cleavage of a carbon–carbon bond in the presence of DIB and a Lewis acid as the halogen source, and thus this method ...

1H NMR metabolomics and lipidomics analysis of neutrophils reveals biomarkers of ageing, inflammageing and frailty

open access: yesThe FEBS Journal, EarlyView.
Using 1H NMR metabolomics, we measured polar and lipid metabolites from human blood neutrophils from people with frailty (n = 31, mean age 84Y), people with rheumatoid arthritis (n = 16, mean age 55Y), robust older (n = 24, mean age 66Y) and healthy younger people (n = 21, mean age 22Y).

Towards an integrated molecular understanding of plant hormones

open access: yesJournal of Integrative Plant Biology, EarlyView.
This review offers a comprehensive overview of the nine plant hormones, delving into their biosynthesis, transport, signaling and crosstalk mechanisms. Because the complexity of plant hormonal control goes beyond these core elements, additional specific features are discussed. Lastly, this review highlights how fundamental insights drive hormonal‐based

Synthesis of 4-styrylquinazolines enabled by a hypervalent iodine-promoted oxidative amination cascade

open access: yesTetrahedron Chem
Efficient synthetic routes to pharmaceutically important 4-styrylquinazolines scaffolds are of considerable interest. Herein, we report a concise synthesis of 4-styrylquinazoline derivatives via a hypervalent iodine-promoted oxidative amination cascade ...
