Design, synthesis, and evaluation of new thiadiazole-based direct inhibitors of enoyl acyl carrier protein reductase (InhA) for the treatment of tuberculosis.

Journal of Medicinal Chemistry, 2015
Mycobacterial enoyl acyl carrier protein reductase (InhA) is a clinically validated target for the treatment of tuberculosis infections, a disease that still causes the death of at least a million people annually.

Utilization of Enzymatically Phosphopantetheinylated Acyl Carrier Proteins and Acetyl−Acyl Carrier Proteins by the Actinorhodin Polyketide Synthase

Biochemistry, 1997
The functional reconstitution of two purified proteins of an aromatic polyketide synthase pathway, the acyl carrier protein (ACP) and holo-ACP synthase (ACPS), is described. Holo-ACPs were enzymatically synthesized from coenzyme A and apo-ACPs using Escherichia coli ACPS.

Enhanced production of branched‐chain fatty acids by replacing β‐ketoacyl‐(acyl‐carrier‐protein) synthase III (FabH)

Biotechnology and Bioengineering, 2015
Branched‐chain fatty acids (BCFAs) are important precursors for the production of advanced biofuels with improved cold‐flow properties. Previous efforts in engineering type II fatty acid synthase (FAS) for BCFA production suffered from low titers and/or ...

Interactions of the acyl chain with the Saccharomyces cerevisiae acyl carrier protein.

Biochemistry, 2015
Acyl carrier protein (ACP) domains are critical integral components of multifunctional type I fatty acid synthases (FAS I) and polyketide synthases (PKSs), where they shuttle the growing adducts of the synthesis between the catalytic domains. In contrast

Chemical and Posttranslational Modification of Escherichia coli Acyl Carrier Protein for Preparation of Dansyl-Acyl Carrier Proteins

Protein Expression and Purification, 2000
Escherichia coli acyl carrier protein (ACP) contains a single tyrosine residue at position 71. The combined o-nitration of apo-ACP Y71 by tetranitromethane and reduction to 3-aminotyrosyl-apo-ACP were performed to introduce a specific site for attachment of a dansyl fluorescent label.

Behavior of acyl carrier proteins on western blots

Biochimica et Biophysica Acta (BBA) - Lipids and Lipid Metabolism, 1990
Acyl carrier proteins (ACPs) from Escherichia coli and Euglena were analyzed on Western blots using rabbit antibodies raised against E. coli ACP. Euglena ACP, unlike that from E. coli, behaves upon electrophoresis under denaturing conditions as its size would predict.

The chain-flipping mechanism of ACP (acyl carrier protein)-dependent enzymes appears universal.

Biochemical Journal, 2014
ACPs (acyl carrier proteins) play essential roles in the synthesis of fatty acids, polyketides and non-ribosomal polypeptides. ACP function requires the modification of the protein by attachment of 4'-phosphopantetheine to a conserved serine residue. The

Mapping a ketosynthase:acyl carrier protein binding interface via unnatural amino acid-mediated photo-cross-linking.

Biochemistry, 2014
Probing and interrogating protein interactions that involve acyl carrier proteins (ACP's) in fatty acid synthases and polyketide synthases are critical to understanding the molecular basis for the programmed assembly of complex natural products. Here, we

An off-pathway folding intermediate of an acyl carrier protein domain coexists with the folded and unfolded states under native conditions.

Angewandte Chemie, 2014
A protein can exist in multiple states under native conditions and those states with low populations are often critical to biological function and self-assembly.
