Loss of ADAMTS5 promotes vascular calcification via versican/integrin β1/FAK signal.
AtherosclerosisINTRODUCTION Extracellular matrix (ECM) proteases have been closely linked to the pathogenesis of vascular calcification. A disintegrin and metalloprotease with thrombospondin motifs-5 (ADAMTS5) is an ECM-degrading enzyme involved in ECM remodeling ...

Computational Insights into ADAMTS4, ADAMTS5 and MMP13 Inhibitor Selectivity
Molecular Informatics, 2012AbstractThe results obtained by means of Molecular Dynamics simulations and Multiway Explorative Data Analysis on ADAMTS4, ADAMTS5 and MMP13 complexed with Marimastat and two cis‐1(S)2(R)‐amino‐2‐indanol ligands suggest that determinant characteristics for ligand binding and selectivity among the three enzymes are to be found in the different protein ...

ADAMTS5 is required for normal trabeculated bone development in the mandibular condyle
Osteoarthritis and Cartilage, 2021Determine the role of the extracellular matrix protease ADAMTS5 in development of the trabeculated bone of the mandibular condyle. The mandibular condyles of wild type and mice deficient in the protease ADAMTS5 were examined for histopathology with Safranin O staining.

Role of ADAMTS5 in Unexplained Fetal Growth Restriction (FGR)
Fetal and Pediatric Pathology, 2016We aim to determine the role of serum and placental A disintegrin and metalloproteinase with thrombospondin motif 5 (ADAMTS5) in fetal growth restriction (FGR).43 pregnancies suffering FGR and 45 healthy ones were homogenized for their body mass indices, ages, and gestational weeks.

Previous studies have indicated the important roles of ADAMTS5 in intervertebral disc degeneration (IDD). However, the mechanisms that regulate ADAMTS5 expression in nuclear pulposus (NP) cells remain largely unknown. Evidence suggests that intergenic transcription may be associated with genes that encode transcriptional regulators. Here, we identified

ADAMTS5 is the major aggrecanase in mouse cartilage in vivo and in vitro
Nature, 2005Aggrecan is the major proteoglycan in cartilage, endowing this tissue with the unique capacity to bear load and resist compression. In arthritic cartilage, aggrecan is degraded by one or more 'aggrecanases' from the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) family of proteinases.

Objective. To identify a biomarker for prediction of the response to infliximab (IFX) in patients with rheumatoid arthritis (RA), we focused on a disintegrin and metalloproteinase with thrombospondin motifs 5 (ADAMTS5) that seems to play a key role in aggrecan degradation in cartilage.Methods.Seventy-three randomly selected patients with active RA were ...

Identification of ADAMTS5 as APP‐Cleaving Enzyme at the APP669 Site
The FASEB JournalCerebral amyloid‐β (Aβ) deposition is a pathological hallmark of the earliest phases of Alzheimer disease (AD). We previously reported APP669‐711 as a novel Aβ‐related peptide detectable in human plasma and developed a composite biomarker that combines ...
