5‐thiohistidine N‐acetyltransferase from Proteiniphilum saccharofermentans
Ovothiol A is a 5‐thiohistidine derivative biosynthesized by a broad range of prokaryotic and eukaryotic organisms. Its redox‐active mercaptoimidazole side chain is believed to protect cells from oxidative stress. The three enzymes that produce ovothiol A from histidine, cysteine, and S‐adenosyl methionine have been identified and characterized.

The protein LmrR is a versatile scaffold to design artificial metalloenzymes. Here, we show that the M8D/A92E (DE) mutations that result in a much more efficient catalyst, destabilize the protein dimerization interface and alter the conformation landscape when binding metal cofactor and substrates are shown.

A series of all‐cis ring fluorinated cyclohexylalanines with progressively increasing levels of vicinal fluorines, as well as 4‐fluorophenylalanine and pentafluoroarylphenylalanine were introduced into the WKYMVm peptide in place of its tyrosine residue, for assays against the G‐protein coupled formylpeptide receptor, FPR2.
