A light‐triggered Time‐Resolved X‐ray Solution Scattering (TR‐XSS) workflow with application to protein conformational dynamics

open access: yesFEBS Open Bio, EarlyView.
Time‐resolved X‐ray solution scattering captures how proteins change shape in real time under near‐native conditions. This article presents a practical workflow for light‐triggered TR‐XSS experiments, from data collection to structural refinement. Using a calcium‐transporting membrane protein as an example, the approach can be broadly applied to study ...

Development of human monoclonal antibodies against TARM1 by yeast display

open access: yesFEBS Open Bio, EarlyView.
Human monoclonal antibodies against TARM1 are generated by yeast display‐guided selection. These antibodies bind to soluble and cell‐surface forms of TARM1. Also, these antibodies exhibit agonistic activity in the NFAT‐GFP reporter assay, indicating that TARM1 signaling can be functionally modulated by antibodies and suggesting TARM1 as a potential ...

Establishing an assay to evaluate d‐amino acid oxidase enzyme kinetics and inhibition using WST‐8 redox dye

open access: yesFEBS Open Bio, EarlyView.
This study investigated a novel WST‐8‐based assay for evaluating d‐Amino acid oxidase (DAO) inhibitors. We confirmed its effectiveness using known inhibitors and found that uremic toxins possess relatively weak inhibitory activity compared to existing drugs.

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

The structural anchoring of spontaneous analogies

open access: yes, 2020
Analogy is a crucial mechanism allowing one to interpret a new situation in the light of passed experiences. The present dissertation seeks to give this statement back its rightful strength by highlighting the ability to perceive as essentially similar experiences showing a different appearance.

Anchored down in Anchorage: Syllable structure and segmental anchoring in French

open access: yes, 2006
We examined tonal alignment and scaling patterns of the start and end points of the French late intonational rise in two read speech corpora. Our goals were twofold. First, we examined several competing hypotheses for characterizing the late rise: 1. an autosegmental-metrical account in which the rise is a bitonal pitch accent composed of a H* pitch ...

Screening and epitope characterization of Nidogen‐2‐specific nanobodies

open access: yesFEBS Open Bio, EarlyView.
Camel immunization and phage display were employed to generate high‐affinity VHH nanobodies against Nidogen‐2. After library construction, biopanning, ELISA screening, sequencing, and recombinant expression, selected nanobodies were purified and characterized, leading to the preliminary exploration of a nanobody‐based sandwich ELISA for specific ...

Three-dimensional stability of a fill slope reinforced by a frame beam anchor plate

open access: yesScientific Reports
The frame beam anchor plate is a new supporting structure reinforcing fill slopes. At present, only a few studies have evaluated its static stability based on the two-dimensional plane assumption. In this paper, the limit analysis method is combined with

Comparison of Pigtail with J Anchor Bolt in Normal Concrete

open access: yesMATEC Web of Conferences, 2018
Anchor bolts have been used to attach the steel structure of concrete and transfer load into the concrete. Anchor bolts in concrete are to withstand the shear force as they connect steel beams to the reinforced concrete foundations.

Decreased cold‐sensing function of the transient receptor potential channel TRPM8 from tailed amphibians

open access: yesFEBS Open Bio, EarlyView.
Despite frogs avoiding low temperatures, examination of four salamander species revealed that none avoided cold and all possessed cold tolerance. Functional analysis of TRPM8, a cold sensor, showed that all salamander TRPM8s had lost their cold sensitivity.
