Angiopoietin-Like Protein 3 (ANGPTL3) Modulates Lipoprotein Metabolism and Dyslipidemia

open access: yesInternational Journal of Molecular Sciences, 2021
Dyslipidemia is characterized by increasing plasma levels of low-density lipoprotein-cholesterol (LDL-C), triglycerides (TGs) and TG-rich lipoproteins (TGRLs) and is a major risk factor for the development of atherosclerotic cardiovascular disorders ...

Angiopoietin-like protein 3 (ANGPTL3) deficiency and familial combined hypolipidemia [PDF]

open access: yesJournal of Biomedical Research, 2019
Three members of the angiopoietin-like (ANGPTL) protein family-ANGPTL3, ANGPTL4 and ANGPTL8- are important regulators of plasma lipoproteins. They inhibit the enzyme lipoprotein lipase, which plays a key role in the intravascular lipolysis of ...

Regulation of lipoprotein metabolism by ANGPTL3, ANGPTL4, and ANGPTL8

American Journal of Physiology-Endocrinology and Metabolism, 2021
Triglyceride-rich lipoproteins deliver fatty acids to tissues for oxidation and for storage. Release of fatty acids from circulating lipoprotein triglycerides is carried out by lipoprotein lipase (LPL), thus LPL serves as a critical gatekeeper of fatty acid uptake into tissues.

Statin therapy reduces plasma angiopoietin-like 3 (ANGPTL3) concentrations in hypercholesterolemic patients via reduced liver X receptor (LXR) activation

open access: yesAtherosclerosis, 2020
Background and aims: Statins suppress hepatic mRNA expression of ANGPTL3 encoding angiopoietin-like 3 in healthy subjects, but it is unknown if plasma ANGPTL3 concentrations are affected by statins prescribed to hypercholesterolemic patients in clinical ...

ANGPTL3: a novel biomarker and promising therapeutic target

Journal of Drug Targeting, 2019
Angiopoietin-like protein 3 (ANGPTL3) belongs to a multifunctional secreted protein that mainly expresses in the liver, and is regulated by numerous post-translational modifications, including multiple cleavage and glycosylation. Accumulating evidences have revealed that ANGPTL3 plays a critical role in both biological processes, such as lipid ...

Angptl3 regulates lipid metabolism in mice

Nature Genetics, 2002
The KK obese mouse is moderately obese and has abnormally high levels of plasma insulin (hyperinsulinemia), glucose (hyperglycemia) and lipids (hyperlipidemia). In one strain (KK/San), we observed abnormally low plasma lipid levels (hypolipidemia). This mutant phenotype is inherited recessively as a mendelian trait.

ANGPTL3 orchestrates hepatic fructose sensing and metabolism

open access: yesCell Reports
Fructose metabolism is linked to metabolic dysfunction-associated steatotic liver disease (MASLD), but the regulatory mechanisms governing fructose uptake remain poorly understood. Here, we demonstrate that MASLD livers exhibit increased uptake of fructose-derived carbons compared to healthy livers and identify that the MASLD hepatocyte secretome can ...

Factoring in ANGPTL3 When LDL Is Refractory

New England Journal of Medicine, 2020
Patients with familial hypercholesterolemia, who have a severely increased low-density lipoprotein (LDL) cholesterol level from birth and are at high risk for premature cardiovascular disease, have...

ANGPTL3 inhibition, dyslipidemia, and cardiovascular diseases

Trends in Cardiovascular Medicine
Optimal management of low-density lipoprotein cholesterol (LDL-C) is a central tenet in the primary and secondary prevention of atherosclerotic cardiovascular disease (ASCVD). However, significant residual cardiovascular risk remains despite achieving guideline-directed LDL-C levels, in part due to mixed hyperlipidemia with elevated fasting and non ...

ANGPTL3 Inhibition in Homozygous Familial Hypercholesterolemia

New England Journal of Medicine, 2017
Evinacumab, a monoclonal antibody that blocks ANGPTL3, was administered to nine adults with homozygous familial hypercholesterolemia. At 4 weeks, LDL cholesterol was reduced by a mean of 49%, with a mean absolute change from baseline of −157 mg per deciliter.
