Three important aspects of immunoglobulin gene organization and structure have emerged from studies of cloned immunoglobulin kappa chain genes. (i) Multiple variable genes are encoded separately in the genome of both immunoglobulin-producing and uncommitted (embryonic) cells, thereby establishing the evolutionary base for generating immunoglobulin ...

The generation of antibody diversity
American Journal of Hematology, 1982AbstractBy their nature, antibody molecules exhibit a wide range of binding specificities. The antigen‐binding properties of the antibody reside entirely in the amino‐terminal portion of the molecule, termed the variable domain. Structurally, the combining site specificity is determined by the amino‐acid residues within 6 short lengths, 3 each in the ...

Antibody Multispecificity Mediated by Conformational Diversity
A single antibody was shown to adopt different binding-site conformations and thereby bind unrelated antigens. Analysis by both x-ray crystallography and pre–steady-state kinetics revealed an equilibrium between different preexisting isomers, one of which possessed a promiscuous, low-affinity binding site for aromatic ligands, including the immunizing ...

The generation of antibody diversity in the turtle
The Journal of Immunology, 1996Abstract The Ab response in reptiles has been studied at the protein level, and in turtles some aspects resemble those of cold-blooded vertebrates from other classes. The genetic bases for these features are not clear. The present study is the first on the IgH organization and complexity of a reptilian Ig gene system.

Somatic Generation of Antibody Diversity
Nature, 1983In the genome of a germ-line cell, the genetic information for an immunoglobulin polypeptide chain is contained in multiple gene segments scattered along a chromosome. During the development of bone marrow-derived lymphocytes, these gene segments are assembled by recombination which leads to the formation of a complete gene.

Generation of antibody diversity in rabbits
Current Opinion in Immunology, 1997Most rabbit B lymphocytes use the same VH gene in V(D)J gene rearrangements and undergo somatic diversification by gene conversion and hypermutation. Recent experiments have shown that V(D)J genes in essentially all rabbit B cells diversify shortly after birth and that this diversification occurs in the gut-associated lymphoid tissue.
