This study reports the development of a fully humanized bispecific T cell engager targeting IL13RA2, a tumor‐associated antigen enriched in glioblastoma. This off‐the‐shelf immunotherapy drives potent, antigen‐dependent T cell activation and tumor killing, and prolongs survival in experimental GBM and other solid tumors models without detectable off ...

An mRNA‐liposome vaccine via dissolving microneedles (ML‐DMN) enables efficient skin‐targeted delivery while preserving mRNA integrity and allowing sustained antigen expression. Unlike intramuscular (IM) injection, ML‐DMN engages skin‐resident immune cells, activates antigen‐presenting cells (APCs), and induces strong T cell and cytokine responses at ...

BackgroundT (8; 21) acute myeloid leukemia (AML) is a special type of acute leukemia, and exhibits a heterogeneous prognosis, with a long-term relapse rate of about 40%. Once t(8; 21) AML patients experience relapse, they have an extremely poor prognosis,

Anti-CD19 chimeric antigen receptor targeting of CD19 + acute myeloid leukemia
Aberrant expression of CD19 in acute myeloid leukemia (AML) is commonly associated with t(8;21)(q22;q22), although AML cases lacking this translocation occasionally express CD19. Mixed-phenotype acute leukemia also frequently expresses CD19. Chimeric antigen receptor (CAR) technology is a major breakthrough for cancer treatment, with the recent ...

This review surveys nanoparticle‐based strategies to enhance adoptive cell therapy, particularly CAR‐T cell approaches, in solid tumor treatment. It describes how nanoparticles can improve tumor immunogenicity and T‐cell infiltration while reducing toxicity, and how they enable in vivo CAR‐T cell generation.

ABSTRACT Despite the transformative impact of cancer immunotherapies such as immune checkpoint blockade, durable clinical responses remain limited. Increasing evidence indicates that antitumor immunity is governed not only by the tumor microenvironment, but also by systemic immune regulation mediated by peripheral immune organs. Among these, the spleen

Truncated form of human CD19 antigen as a suicide gene to control T-cell alloreactivity: ΔCD19
Donor lymphocyte infusion (DLI) is employed to either treat or prevent relapse in patients with hematologic malignancies, as well as to accelerate recovery of adaptive immunity, after allogeneic hematopoietic stem cell transplantation (allo-HSCT). With the increased use of DLI, there is renewed interest in the development of approaches able to prevent ...

Sono‐Mechanogenetics: Linking Ultrasound Physics With Cellular Mechanobiology
Sono‐mechanogenetics links ultrasound physics with cellular mechanotransduction to enable noninvasive control of engineered biological systems. Acoustic forces generate distinct deformation modes that activate intracellular signaling pathways, which can be coupled to synthetic gene circuits to regulate diverse cellular functions, including gene ...
