Rolig, Peng, and colleagues have shed new light on how dual blockade of PD1 and LAG3 enhances antitumor immunity. They found that response to immunotherapy with anti‐PD1 + anti‐LAG3 was associated with reprogramming of canonically suppressive CD4+ regulatory T cells to an inflammatory state.

Antitumor Activity of Cytogenin.
Antitumor effect of cytogenin against IMC carcinoma in mice was investigated. Since cytogenin did not show cytotoxicity against tumor cells in vitro at 50 micrograms/ml and toxicity at more than 2,000 mg/kg i.p., it was considered that the antitumor effect is due to host mediated events.

Chimeric diphtheria toxin–CCL8 cytotoxic peptide for breast cancer management
DTCCL8 is a recombinant fusion toxin that targets cancer cells expressing chemokine receptors. By combining diphtheria toxin with CCL8, DTCCL8 binds to multiple receptors on tumor cells and induces selective cytotoxicity. This strategy enables receptor‐mediated targeting of cancer and may support the development of chemokine‐guided therapeutics ...

ANTITUMOR ACTIVITY OF ECHINOSPORIN
Echinosporin isolated from a Streptomyces culture showed antitumor activity against rodent tumor models such as leukemia P388, P388/VCR, and fibrosarcoma Meth 1. It was marginally active against melanoma B16 and sarcoma 180. It was not active against Lewis lung carcinoma and xenograft MX-1. It inhibited the colony formation of HeLa S3 cells with a wide

Raphin‐1 reduces the survival of PED‐DHGG cells and enhances their radiation sensitivity through both PeIF2α‐dependent and PeIF2α‐independent mechanisms. Raphin‐1 sustains elevated levels of PeIF2α, contributing to its PeIF2α‐dependent effects. Additionally, raphin‐1 interacts with CReP to mediate a separate radiosensitizing pathway that operates ...

Inhibition of protein SUMOylation has been shown to block tumorigenesis; however, the specific mechanisms by which SUMOylation controls the tumor‐initiating capacities remain elusive. Li et al. describe the role of Etv1 SUMOylation in cancer stem cells using mouse models of mammary gland tumorigenesis.

In prostate carcinoma, lactic acid, secreted by highly glycolytic cancer‐associated fibroblasts, is imported into tumor cells through the MCT1 transporter and prevents RSL3 and erastin‐induced ferroptosis (A). Targeting of carbonic anhydrase IX/XII, the main extracellular pH regulators, in tumor and stromal cells reduces microenvironmental acidosis and
