ESCs were first derived from mouse diapause blastocysts or isolated ICMs plated onto fibroblast feeders in a medium containing FBS. Feeders were then replaced with LIF, and later, BMP4 was substituted for serum. GSK3 and ERK inhibition finally enabled efficient derivation of ESCs from embryos of all strains of mice and rats.
