Molecular structure and function of the porcine arachidonate 12-lipoxygenase gene.

The gene encoding arachidonate 12-lipoxygenase was cloned from a porcine EMBL3 genomic library using a cDNA probe of the enzyme, and its nucleotide sequence was determined. The gene consists of 14 exons with 13 introns, and spans approximately 8 kilobases.

In thoracic aortic dissection (TAD), elevated levels of the arachidonic acid metabolite 12‐HETE, produced by macrophage‐specific 12/15‐lipoxygenase (12/15‐LOX), bind to the BLT2 receptor on vascular cells. This activates the NOX‐1/ROS/NF‐κB signaling pathway, leading to IL‐6 release.
