Novel (S)‐5′‐guanidinopropyl uridine analogs bearing 2′‐O‐methyl or 2′‐fluoro substitutions enhance nuclease resistance and mismatch discrimination while preserving A‐form RNA structure and potent RNAi activity. These modifications provide a promising strategy for improving the stability, specificity, and therapeutic potential of siRNA‐based medicines.

Panoptes as an example for an antiphage system consists of OptS producing a cyclic dinucleotide that prevents activation of OptE, a transmembrane protein.
