Mitochondria‐associated membranes (MAMs): molecular organization, cellular functions, and their role in health and disease

open access: yesFEBS Open Bio, EarlyView.
Mitochondria‐associated membranes (MAMs) are contact sites between the endoplasmic reticulum and mitochondria that regulate calcium signaling, lipid metabolism, autophagy, and stress responses. This review outlines their molecular organization, roles in cellular homeostasis, and how dysfunction drives neurodegeneration, metabolic disease, cancer, and ...

Production of antibacterial compounds by a Steely hybrid polyketide synthase in Dictyostelium

open access: yesFEBS Open Bio, EarlyView.
By selecting appropriate production species under optimised culture conditions, new chlorinated dibenzofuran compounds, which previously could not be analysed in sufficient quantities, were identified from cellular slime mould fruiting bodies. The strong antibacterial activity exhibited by these compounds suggests a link to the ecological function of ...

The mitochondrial ATP synthase is a shared drug target for aging and dementia

open access: yesAging Cell, 2018
Aging is a major driving force underlying dementia, such as that caused by Alzheimer's disease (AD). While the idea of targeting aging as a therapeutic strategy is not new, it remains unclear how closely aging and age‐associated diseases are coupled at ...

HSP70 governs permeability and mechanotransduction in primary human endothelial cells

open access: yesFEBS Open Bio, EarlyView.
HSP70 chemical inhibition reduces endothelial cell proliferation and increases permeability, the latter supported by normal interendothelial junctional protein distribution. HSP70 also plays a role in shear stress response, a hemodynamic force naturally present in blood vessels and correlated with vessel protection.

Parkinson’s disease protein DJ-1 regulates ATP synthase protein components to increase neuronal process outgrowth

open access: yesCell Death and Disease, 2019
Familial Parkinson’s disease (PD) protein DJ-1 mutations are linked to early onset PD. We have found that DJ-1 binds directly to the F1FO ATP synthase β subunit.

Molecular determinants of signal transduction in tropomyosin receptor kinases

open access: yesFEBS Open Bio, EarlyView.
Tropomyosin receptor kinases control critical neuronal functions, but how do the same receptors produce diverse cellular responses? This review explores the structural mechanisms behind Trk signaling diversity, focusing on allosteric modulation and ligand bias.

Analyzing conformational changes in single FRET-labeled A1 parts of archaeal A1AO-ATP synthase

open access: yes, 2018
ATP synthases utilize a proton motive force to synthesize ATP. In reverse, these membrane-embedded enzymes can also hydrolyze ATP to pump protons over the membrane.

Evolutionary dynamics of the chloroplast genome in Daphne (Thymelaeaceae): comparative analysis with related genera and insights into phylogenetics

open access: yesFEBS Open Bio, EarlyView.
Comparative analysis of chloroplast genomes from 14 genera of Thymelaeaceae revealed variation in gene content, ranging from 128 to 142 genes, primarily influenced by IR expansion/contraction events and pseudogenization of ndhF, ndhI, and ndhG. Two large inversions were detected within the large single‐copy region, including a synapomorphic inversion ...

Structure and conformational states of the bovine mitochondrial ATP synthase by cryo-EM

open access: yeseLife, 2015
Adenosine triphosphate (ATP), the chemical energy currency of biology, is synthesized in eukaryotic cells primarily by the mitochondrial ATP synthase. ATP synthases operate by a rotary catalytic mechanism where proton translocation through the membrane ...
