Tau acetylation at K331 has limited impact on tau pathology in vivo

open access: yesFEBS Letters, EarlyView.
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

Calpain small subunit homodimerization is robust and calcium‐independent

open access: yesFEBS Letters, EarlyView.
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

Multi‑objective Feature Selection Algorithm for Neighborhood Rough Set Under Mixed Hierarchical Dependence

open access: yesShuju Caiji Yu Chuli
Accuracy and efficiency are the key metrics for evaluating the performance of feature selection algorithms. They correspond to the attribute dependence and reduction scale of neighborhood rough sets respectively. Conventional feature selection algorithms

A methionine‐lined active site governs carbocation stabilization and product specificity in a bacterial terpene synthase

open access: yesFEBS Letters, EarlyView.
This study reveals a unique active site enriched in methionine residues and demonstrates that these residues play a critical role by stabilizing carbocation intermediates through novel sulfur–cation interactions. Structure‐guided mutagenesis further revealed variants with significantly altered product profiles, enhancing pseudopterosin formation. These

Protecting built property against fire disasters: Multi ‐attribute decision making with respect to fire risk

open access: yesInternational Journal of Strategic Property Management, 2010
The protection of buildings against fire disasters may require a comparison of alternative fire safety designs. The fire safety solutions can be compared by means of a general methodology known as multi‐attribute selection or multi‐criteria decision ...

Improving Lazy Attribute Selection

open access: yesJournal of Information and Data Management, 2011
Attribute selection is a data preprocessing step which aims at identifying relevant attributes for a target data mining task - specifically in this article, the classification task. Previously, we have proposed a new attribute selection strategy - based on a lazy learning approach - which postpones the identification of relevant attributes until an ...

Cytoplasmic p21 promotes stemness of colon cancer cells via activation of the NFκB pathway

open access: yesMolecular Oncology, EarlyView.
Cytoplasmic p21 promotes colorectal cancer stem cell (CSC) features by destabilizing the NFκB–IκB complex, activating NFκB signaling, and upregulating BCL‐xL and COX2. In contrast to nuclear p21, cytoplasmic p21 enhances spheroid formation and stemness transcription factor CD133.

Suite of decision tree-based classification algorithms on cancer gene expression data

open access: yesEgyptian Informatics Journal, 2011
One of the major challenges in microarray analysis, especially in cancer gene expression profiles, is to determine genes or groups of genes that are highly expressed in cancer cells but not in normal cells. Supervised machine learning techniques are used
