Mepolizumab Modifies Blood Eosinophil Proteome and Transcriptome in Severe Eosinophilic Asthma

open access: yesAllergy, EarlyView.
This study investigates the effect of mepolizumab on the proteomic and transcriptomic profile of blood eosinophils in SEA patients. Mepolizumab reduces the migratory capacity and effector‐related molecular signatures of circulating eosinophils in SEA patients.

Failed Mucosal Allostasis Distinguishes Murine IgG‐ From IgE‐Mediated Recurrent Severe Anaphylaxis

open access: yesAllergy, EarlyView.
This study investigates IgE‐ and IgG‐mediated pathways in acute and recurrent anaphylaxis using a murine ASA‐PSA model, assessing barrier integrity, immune activation and tissue remodelling across pulmonary and intestinal compartments. Recurrent IgE‐ and IgG‐mediated anaphylaxis produces divergent mucosal outcomes despite comparable systemic severity ...

Pre‐Seasonal Treatment With Recombinant Carrier‐Based Birch Vaccine Improves Birch Pollen Allergy

open access: yesAllergy, EarlyView.
We conducted a two stage SCIT trial with GNR‐127 containing a recombinant fusion protein consisting of Bet v 1 and Mal d 1‐derived peptides fused to PreS from HBV. Stage 1 identified two safe maximum tolerated doses (40 μg and 80 μg). In the following Stage 2, five preseasonal injections were well tolerated, induced dose‐dependent allergen‐specific IgG,

IgE‐Binding Bacterial Components Not Yet Documented in the Allergen Databases and Their Allergenic Mechanisms: An EAACI Position Paper

open access: yesAllergy, EarlyView.
ABSTRACT Among bacteria that are associated with the development and/or exacerbation of allergic/atopic diseases, several have been described to produce IgE‐binding components. However, only one bacterial protein, Bac s 1, is currently listed in the official database of the World Health Organization/International Union of Immunological Societies (WHO ...

Understanding the Type 2 Immunity: An Evolving View Beyond Allergic Diseases

open access: yesAllergy, EarlyView.
ABSTRACT Type 2 (T2) immunity is classically associated with defense against helminths and environmental threats, from a physiological perspective, and with allergies and eosinophilic inflammatory diseases, in a pathological sense. However, growing evidence reveals that T2 pathways also support other essential homeostatic functions, including tissue ...

Single‐Cell Profiling Links T‐Cell Accumulation and TH2/TH17 Dynamics to Eosinophilic Esophagitis (EoE) Activity in Children

open access: yesAllergy, EarlyView.
Active EoE is associated with more and deeper epithelial eosinophil, mast cell, and T cell infiltration; inactive EoE resembles controls. During remission, PPI treatment leaves more tissue resident CD3+ T‐cells than TS. In active EoE, TH2 cells, IL‐10hi Tregs, and CD8+ cytotoxic T cells are enriched, while TH17 cells are relatively depleted.

Diagnostic Performance of Recombinant Peanut Allergens in Children: sIgE Cut‐Offs and Anaphylaxis Risk

open access: yesActa Paediatrica, EarlyView.
Diagnostic performance of recombinant peanut allergens for anaphylaxis risk in children. (A) Study population and clinical group. (B) Component‐specific IgE cut‐offs and associated odds ratios. (C) Forest plot of odds ratios for anaphylaxis. (D) Clinical interpretation of component‐specific IgE cut‐offs. ABSTRACT Aim Component‐resolved diagnostics (CRD)

Factors Associated With Omalizumab Response in Chronic Spontaneous Urticaria: Are There Age‐Related Differences?

open access: yesActa Paediatrica, EarlyView.
ABSTRACT Aim This study aimed to explore clinical and laboratory markers of omalizumab response in paediatric and adult chronic spontaneous urticaria (CSU) patients. Methods The study included 32 children and 67 adults with CSU receiving omalizumab, and 29 paediatric and 30 adult healthy controls; associations with treatment response were evaluated ...
