Prediabetes Associates With Musculoskeletal Alterations Independent of Total Body Adiposity
ABSTRACT Background Excess adiposity is a major risk factor for insulin resistance, prediabetes, and Type 2 diabetes and increases the risk for sarcopenia and osteosarcopenia later in life. It has been proposed that altered metabolic function and musculoskeletal status in people with obesity are directly linked, presumably because they share common ...

ABSTRACT Background Skeletal muscle wasting and weakness are prominent disease features. Originally considered to arise from common transcriptional changes, recent analyses demonstrated that different stimuli induce muscle wasting via largely distinct mRNA and protein changes.

Progress in RNA‐Targeted Therapeutics for Human Diseases
RNA‐targeted therapies are revolutionizing molecular medicine by transitioning from a “protein‐centric” focus to an “RNA‐regulatory network” approach. Leveraging RNA's diverse roles in gene regulation, signaling, and epigenetic modifications, advanced platforms such as ASOs, siRNA, miRNA, mRNA, aptamers, shRNA, and CRISPR/Cas systems are enabling ...

VMP1 promotes angiogenesis in glioblastoma (GBM) through VEGFA‐VEGFR2 signaling and activation of endothelial cells. The FDA‐approved anti‐VEGFA antibody, bevacizumab (BEV), can counteract VMP1's tumor‐promoting effect in GBM. Targeting VMP1 or its associated pathways could be a new therapeutic strategy, either alone or in combination with BEV.

RCCD1 is upregulated in colon cancer and cancer‐associated fibroblasts (CAFs). In CAFs, RCCD1 overexpression activates AMPK/mTOR/ULK1 signaling, enhancing autophagy. This stimulates autophagic secretion of WNT5A. WNT5A then activates the nonclassical Wnt/CaMKII/ERK pathway in tumor cells, promoting epithelial‐mesenchymal transition (EMT), proliferation,

Dietary protein restriction protects the aging heart in the context of obesity by limiting mitochondrial DNA leakage and suppressing cGAS–STING‐driven inflammation. Through AMPK–ULK1‐dependent mitophagy, protein restriction restores mitochondrial quality control, reduces cardiac remodeling, and preserves metabolic homeostasis during obesity‐associated ...

Non‐coding RNAs (ncRNAs) regulate breast cancer radioresistance via cell cycle, DNA repair and tumour microenvironment pathways. Targeting ncRNAs (e.g., HOTAIR and miR‐155) with RNA‐based therapies (ASOs and CRISPR) shows promise but faces delivery challenges.

Crystal structure of the Bcl-XL-Beclin 1 peptide complex: Beclin 1 is a novel BH3-only protein.
Bcl-2 family proteins are key regulators of apoptosis and have recently been shown to modulate autophagy. The tumor suppressor Beclin 1 has been proposed to coordinate both apoptosis and autophagy through direct interaction with anti-apoptotic family members Bcl-2 and/or Bcl-X(L).
